Pleurotus geesteranus has been produced with large-scale factory manner,which produces a large amount of P.geesteranus residue every year.In this paper,the design of Plackett Burman(PB)and the analysis of response surface methodology(RSM)were used to optimize the extraction conditions of P.geesteranu residue polysaccharide(PRPS).After being treated with snailase,the enzymatic-extractable P.geesteranus resource polysaccharide(EPRPS)was obtained and used to detect its antioxidant activity in vitro.The effects of EPRPS on the serum indexes of CRF mice induced by adenine and the activities of antioxidant enzymes in renal tissue were analyzed,which could be used as a reference for the screening of natural renal injury drugs.The main results are as follows:(1)The theoretical yield of PRPS was 2.81% when the extraction times were 2 times,the ethanol concentration was 93.06%,and the extraction degree was 87.12 ?.For practical operation,the extraction conditions were adjusted to 90% ethanol concentration,87 ?extraction temperature,theoretical yield was 2.78% and the verification yield of PRPS was2.62 ± 0.12%.(2)At 800 mg/l,the scavenging effects of EPRPS on hydroxyl,superoxide anion and1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)radicals were 58.71 ± 1.71%,43.13 ± 1.62%and 64.89 ± 1.72%,respectively,which was 49.54%,41.13% and 59.08% higher than those of PRPS.The reducing power of EPRPS was 0.48 ± 0.01%,which was 51.16% higher than that of PRPS.(3)The EPRPS could increase the activities of catalase(CAT),glutathione peroxidase(GSH PX)and superoxide dismutase(SOD)in kidney of CRF mice,showing that EPRPS had strong antioxidant capacity.The contents of blood urea nitrogen(BUN),uric acid(UA)and creatine kinase(CK)of serum were decreased,endogenous waste was removed,electrolyte and acid-base balance were maintained.EPRPS could reduce the contents of LPO,TNF-?,IL-6 and nuclear transcription factor in vivo(factor kappaB,NF KB)activity or level,delay glomerulosclerosis and promote functional recovery,and the repaired effect of EPRPS had a dose manner.The results of renal histopathology showed that the intervention of EPRPS could ameliorate the chronic renal injury induced by adenine.EPRPS had anti-oxidation,anti-inflammatory and anti fibrosis activities,indicating that EPRPS might provide a basis for the comprehensive development and utilization of the polysaccharide.
