Carotenoids have a lot of biological activities,such as free radical scavenging and quenching active oxygen.This activity is similar to vitamin E,with astaxanthin being the most active.In this study,a carotenoids over-producing strain of Phaffia rhodozyma was used as experimental material.Extraction,recrystallization,HSCCC and column chromatography was used to purify the carotenoids in P.rhodozyma.In addition,using the DPPH,ABTS+,·OH,FRAP,ORAC and olive oil systems to determine the antioxidant activity and synergistic effects of astaxanthin,?-carotene and echinenone.The main results were as follows:?1?The three substances of isolated and identified from P.rhodozyma were echinenone,?-carotene and astaxanthin respectively.The purity of astaxanthin of recrystallization and HSCCC preparation were calculated to be 83.78%and 93.65%,respectively.The astaxanthin from P.rhodozyma?astaxanthin of recrystallization and astaxanthin of HSCCC preparation?was composed of three cis-trans isomerism,stereoisomeric was mainly?3R,3'R?-astaxanthin.Synthetic astaxanthin was composed of only one cis-trans isomerism,and it had a stereoisomeric ratio of 1:2:1 for the?3S,3'S?,?3S,3'R?,?3R,3'R?.?2?The scavenging activities of DPPH,ABTS+and·OH radicals of astaxanthin of synthetic and astaxanthin of HSCCC preparation were improved with increasing concentrations.Astaxanthin of synthetic had the highest scavenging capacity of ABTS+and·OH radicals,and the EC500 values were 61.21±4.17?g/mL and 26.12±5.14?g/mL,respectively.Echinenone had the strongest scavenging capacity of DPPH radical,and the EC50 value was 19.90±2.72?g/mL.Astaxanthin of synthetic and astaxanthin of HSCCC preparation had ferric ion reducing power,with the astaxanthin of synthetic being the strongest and the FRAP value was 0.2816±0.005mmol/L.Echinenone,?-carotene,astaxanthin of synthetic and astaxanthin of HSCCC preparation possessed some inhibitory effect on oil oxidation,and the inhibitory effect of astaxanthin of synthetic and astaxanthin of HSCCC preparation was significant.Astaxanthin of synthetic with the highest ORAC value,was significantly higher than the other three samples?P<0.05?.?3?The antioxidant activity of two components?astaxanthin,?-carotene or echinenone?showed that the combination scavenging DPPH,ABTS+and·OH radicals of low concentrations of astaxanthin with?-carotene and echinenone,respectively,had the tendency to promote synergistic antioxidant activity.?4?The factor analysis of the synergistic antioxidant among astaxanthin,?-carotene and echinenone showed that the synergistic effect of three components had a significant effect on the interaction of DPPH,ABTS+and·OH radicals antioxidant system.When the mass concentrations of astaxanthin,?-carotene and echinenone were 4,4,8?g/mL,2,4,8?g/m L and 8,8,2?g/mL,respectively,the maximum clearance rates were 48.57%,24.11%and 34.81%,respectively.The experimental results of this paper provide the theoretical basis and technical support for the preparation of carotenoids from P.rhodozyma and the synergistic effect of antioxidant and the study of the composition of astaxanthin from different sources.
