Nanodiamonds(NDs)have excellent physical and chemical properties,abundant and adjustable surface groups and good biocompatibility,which makes them have a wide application prospect in electronic devices,luminescent materials,composites,catalysis and biomedicine.Herein,the nano-diamonds were dispersed and functionalized by mechanical grinding,high-temperature annealing and surface modification,then the Fucoxanthin(Fx)was connected to construct a nano drugloading system ND-Fx to study its activity inhibition effect on Hela cervical cancer cells.First,the parameters of high temperature air oxidation treatment were determined by synchronous thermal analysis test,that is,the temperature was increased to 400°C at a rate of 5°C/min,and starting to cool down after 150 min of heat preservation.The sp2 carbon on the surface of nano-diamonds was reduced after treatment,and the sp3 carbon was revealed.Then the surface modification of nanocrystalline diamond was carried out with chromic acid lotion and strong oxidizing mixed acid,and the carboxylated nanocrystalline diamonds(ND-COOH)with average particle size of 91 nm,surface potential of-35.5 m V was obtained,and the dispersion stability could be maintained for a long time.The ND-COOH were surface modified using sulfoxide chloride to obtain acylchloride nanodiamonds(ND-COCl)with stronger reactivity.The nano-drug-loaded composite particles ND-Fx were prepared by combining ND-COOH,ND-COCl with fucoxanthin,respectively.By the characterization of Fourier transform infrared spectrometer and UV-Vis spectrophotometer,the effective combination of diamond nanoparticles and fucoxanthin was proved,and the loading of ND-Fx prepared by the two methods was calculated,and the content of fucoxanthin reached 19.9% and 24.42%,respectively.The results show that the nano-drug-loaded composite particles ND-Fx with higher drug loading capacity can be obtained by using ND-COCl as the material.The drug release amount of ND-Fx at different p H and different release time was tested.As the p H decreased,the release rate of the drug increased and the release amount increased.the release amount could reach more than 89.62%.Different concentrations of NDs,fucoxanthin,ND-Fx,the mixture of nanodiamonds and fucoxanthin(ND+Fx)were co-cultured with Hela cells for 24 h before MTT test.The results showed that NDs had no negative effect on cell bioactivity,while the inhibitory effect of ND-Fx on Hela cell activity was significantly higher than that of fucoxanthin monomer.
