The synthesis process of a n-butyl 2-cyanoacrylate monomer for medical adhesive is optimized on the basis of the previous solvent-free method.The scheme is applying paraformaldehyde instead of formaldehyde solution under the condition of adding composite additives and cyanide n-butyl acetate to synthesize n-butyl 2-cyanoacrylate via Knoevenagel reaction.Using infrared spectroscopy(FT-IR)and nuclear magnetic resonance(~1H-NMR)characterizated the molecular structure of product.Gas chromatograph,universal mechanical tester and ubbelohde viscometerother machine were used to characterizate the performance of product.The influence of the synthesis reaction on usage amount of the amount of additives,time of dehydrating agent joined system and adjusting different p H after the completion of the reaction were mainly discussed.Experiment results show that the use of composite additives and paraformaldehyde simplifies synthetic process to a certain extent,and with the best use of reactant ratio of paraformaldehyde and n-Butyl cyanoacetate was 1.05:1,the ratio of composite additives and n-Butyl cyanoacetate)was 0.05:1;the dehydrating agent joined the system before the reaction has better yield than joined when the reaction have finished;the monomer reached the highest yield when using phosphoric acid adjusted the system p H between 7.0 and 7.5after the reaction completed system.The Infrared spectroscopy(FT-IR)and nuclear magnetic resonance(~1H-NMR)have characterizated the monomer structure successfully,Optimization of process compared with the traditional synthesis process,the cost was reduced by 10.1 percents,monomer yield increased by 6.8 percents,the antibacterial experiments in vitro confirmed that the monomer can meet preliminary clinical requirements as well.The modification research on ethyl ?-cyanoacrylates used nano silica,hydroxylation multi-walled carbon nanotubes,and the epoxy resin showed the application of nanometer materials improved the viscosity and tensile shear strength significantly.The discovery can broaden the application scope and prospects of this series of adhesive and possess a strategic significance.
