Metabolomics is an emerging and popular technology which is able to make determination simply,quickly and accurately with the characteristics of high throughput and high resolution..It plays an important role in effectively predicting the levels of metabolites in the body,and timely diagnoses the body's physiological and pathological state and guides the clinical intervention.Liquid chromatography-tandem mass spectrometry(LC-MS/MS)is a powerful detection technique for metabolomics,which has now attracted more attention and extended more applications.LC-MS/MS is the "gold standard"for the quantitative determination of endogenous metabolites.Ceramides,as a family of small molecules of metabolism,has been proved to be potential biomarkers in many diseases.Quantification of ceramides are of great significance for predicting disease progression and prognosis.There are multiple members in the ceramide family,but the species of measurements in the previous study are limited.This paper uses the LC-MS/MS platform to develop a broad-spectrum ceramide quantification method,which is currently the method for measuring the most species of ceramides in one injection,and has met all the requirements of the verification of the mass spectrometry detection assessment,which lay the foundation for the study of the role of ceramide in the diseases.This article will provide reference technology for the quantification scheme of ceramide,and provide technical support for the accurate and comprehensive determination of ceramide.European and American research teams have confirmed in the foreign coronary heart disease patient cohort that ceramide levels have an early warning and evaluation effect on the risk of patients' adverse events in the short term,and developed a risk assessment score model,but there are differences in diet and genetics between Chinese and foreign population.In China,such research is still pending.This article aims at the patient population in China,to identify specific species of ceramide as potential biomarkers in patient population,and generate three applicable integral models,which can be used as a scoring system for Chinese patients to improve the capability of predicting cardiovascular events and even death in patients with coronary heart disease.This early risk warning system provides effective screening method for high-risk groups and therefor to improve clinical intervention.
