The yeast β-glucan in the cell wall of Saccharomyces cerevisiae has excellent biological activities,such as enhancing the body’s immunity,anti-tumor,preventing and treating cancer,anti-infection,anti-radiation and anti-oxidation,lowering blood cholesterol and regulating gastrointestinal bacteria Group and other biological activities.However,due to its structural reasons,yeast β-glucan is difficult to dissolve in water and most organic solvents.Improving and improving the solubility of yeast β-glucan in water or other solvents will be beneficial to its use in food,medicine,Application in the field of health care products and skin care products.This paper mainly studies the extraction method of yeast β-glucan from Saccharomyces cerevisiae from three different sources,explores the modified method of the extracted yeastβ-glucan,and explores the quantitative detection method of its content and molecular weight.The yeast β-glucan is extracted by multi-step enzyme alkaline method,and the purity of the product is above 87%,and the impurity protein is below 1%.This method was used to extract yeast β-glucan from three different sources of Saccharomyces cerevisiae respectively.The three Saccharomyces cerevisiae were brewer’s yeast slime,selenium-enriched yeast cell wall and yeast strain SAM.The product quality obtained was good.This method It has certain universality for the extraction of yeast β-glucan.The preferred protease in the extraction method is alkaline protease,and the enzymolysis effect is significantly higher than other proteases.Modification of yeast β-glucan,insert trimethylsilane at the hydroxyl end to shield part of the hydroxyl group,reduce the interaction between intermolecular hydrogen bonds,and prepare ethanol-soluble silylated yeastβ-glucan.Using dichlorodimethylsiloxane as a cross-linking agent,yeastβ-glucan and sodium hyaluronate are cross-linked,and the prepared derivative product has excellent water retention performance.An acetyl group is introduced at the C-2 position of yeast β-glucan to prepare an acetylated yeastβ-glucan that can be dissolved in acetone and oleic acid.The GOPOD method uses specific enzymes to hydrolyze the yeastβ-glucan stepwise,which can accurately detect the yeast β-glucan content.The enzymolysis-liquid phase method uses the hydrolytic enzyme in the GOPGD method to the yeast β-glucan Sugar is hydrolyzed,and then the hydrolysate glucose is measured by HPLC method,and a precise and low-cost content detection method is developed.The molecular weight of the water-soluble yeast β-glucan modified product was detected by gel permeation chromatography.The method has good precision and stability.
