Magnetic molecularly imprinted polymers?MMIP?have attracted more and more attention due to their special selectivity for template molecules and excellent magnetic properties.MMIP can be separated trace target from solution.Therefore,MMIP was used to extract the allocryptopine and protopine from the waste liquid of Macleaya cordata?Willd?R.Br..This method has the characteristics of high efficiency,simplicity,low cost and no pollution to the environment.In recent years,because of its antithrombotic,anti-inflammatory,antibacterial and insect repellent activities,protopine have been widely concerned.Human serum albumin?HSA?is needed as a carrier to deliver drugs to target cells.Therefore,it is of great significance to study the interaction between protopine and human serum albumin in order to elucidate the mechanism of the binding of these alkaloids in human body and their pharmacokinetics.The general method to study the conformational changes of proteins is fluorescence spectroscopy.Fluorescence spectrometry also has the characteristics of high selectivity,high sensitivity,low cost and simple method.Therefore,it is widely used in the study of the interaction between proteins and small molecule drugs.This study mainly studies:?1?Used Fe3O4@Si O2,4-vinylpyridine,ethylene glycol dimethacrylate and protopine as magnetic nucleus,functional monomer,cross-linking agent and template agent,the PRO-MMIP was synthesized.The performance of mmip was studied by adsorption experiment.The experimental results showed that mmip had high selectivity,recoverability and good adsorption performance?21.96 mg/g 298 K?for the PRO.The adsorption process was in accordance with Langmuir isotherm and pseudo second order kinetic model.After six adsorption desorption experiments,the adsorption capacity of MMIP decreased only 7.9%.In the selective adsorption experiment,the adsorption performance of MMIP to the PRO is much greater than that of the substances with similar functional groups.?2?A novel type of magnetic molecularly imprinted polymers?MMIP?as the solid-phase extraction sorbent was prepared,which can extracted effectively the allocryptopine,which was from the waster of Macleaya cordata?Willd?R.Br.In this study,MMIP was synthesized by using Fe3O4@Si O2,4-vinyl-pyridine,ethylene glycol dimethacrylate and allocryptopine,and these ingredients worked as magnetic core,functional monomer,cross-linker and template,respectively.Concluded by the calculation of Gaussian 09 software,different ratio models of 4-vinyl-pyridine and allocryptopine were simulated,and the optimal ratio was 1:5,the energy was-2205.34 k J·mol-1.Transmission electron microscopy,vibration sample magnetometry,X-ray diffraction,fourier transform infrared spectroscopy and thermogravimetric analysis were used to determine the morphology and structural of MMIP.Furthermore,the results of adsorption experiments indicated that MMIP had high selectivity,excellent recyclability and good adsorption performance?9.86 mg/g,298K?.The adsorption process was consistent with the Langmuir adsorption isothermand pseudo-second-order kinetics model.After 6 times adsorption-desorption experiments the adsorption amount of MMIP only reduced to8.5%.In the experiments of selective adsorption,MMIP has better adsorption properties for allocryptopine(ALL,C21H23NO5)than those having the same functional group.The limit of detection?LOD?was 0.4?g/m L.The relative standard deviation ranged from 0.09%to 0.72%.The recovery of allocryptopine in samples ranged from 93.60%to 106.19%.?3?Pro has a strong fluorescence effect on HSA,and with the increase of the concentration of PRO,the endogenous fluorescence of HSA increases gradually,and its synchronous fluorescence also increases gradually.At 300K,310K and 315K,the apparent binding constants?K?of protoopioids to HSA were 0.12,0.026 and 0.12,respectively.These results show that the fluorescence properties of HSA are enhanced by PRO.The blue shift is caused by the amino acids in has in a more hydrophobic environment.The results show that the magnetic molecularly imprinted materials can effectively adsorb allocryptopine and protoopioid bases.It provides a valuable research basis for the extraction and separation of alkaloids from natural products.The main interaction forces between protoopioids and HSA are van der Waals force and hydrogen bond force.
