China owns the most abundant bamboo resources in the world,and it is widely distributed.And it contain a large amount of active ingredients,especially flavonoids.In this study,as one importment species of Phyllostachys,the Phyllostachys edulis leaves were studied;A liquid phase detection method for five major flavone glycosides was established;At the same time,the differences in the content of four flavone C-glycosides in 24 bamboo leaves were compared.It provides a basic study for the rational application of bamboo leaf resources.The research results are as follows:(1)Extracting flavonoids from Phyllostachys edulis leaves with organic solvents extract.Extraction was purified by AB-8 macroporous absorption resin and petroleum ether.And extraction was purified by ODS reversed phase column again,then extraction by 10% ethanol,15% ethanol,20% ethanol,25% ethanol,30% ethanol and 25% ethanol eluted respectively.The elutions were isolated by Sephadex LH-20 and Sepacore low-pressure HPLC Separate system.Six compounds were isolated from Phyllostachys edulis leaves.On the basis of physic-chemical characters and spectroscopic analysis such as NMR and MS,6 compounds were identified as: Isoorientin(1),Orientin(2),Vitexin(3),Isovitexin(4),Luteolin(5),Luteoloside(6).(2)The HPLC separation was performed on an Inertsil ODS-3(250 mm×4.6 mm,5 ?m)column with the mobile phase of acetonitrile(B)and 0.5% phosphoric acid in water(C)with gradient elution at a flow rate of 1 m L/min..The column temperature was 30?,and the detection wavelength was 270 nm.The detection of five flavone glycosides was achieved.The content of isoorientin,orientin,vitexin,isovitexin and luteoloside in the powder samples of bamboo leaf flavonoid extract were 14.76,2.82,4.24,22.32 and 2.78 mg/g,respectively.(3)The HPLC separation was performed on an Inertsil ODS-3(250 mm×4.6 mm,5 ?m)column with the mobile phase of acetonitrile(B)and 0.5% phosphoric acid in water(C)with gradient elution at a flow rate of 1 m L/min.The column temperature was 30 ?,and the detected wavelength was 340 nm.The difference of four flavone C-glycosides in 24 bamboo leaves of Phyllostachys and P.propinqua in the four seasons was analyzed by one-way analysis of variance.At the same time,24 bamboo leaves of Phyllostachys of the same sampling area were clustered according to the difference of four kinds of flavonoids.The results showed big differences in flavone C-glycosides content among bamboo leaves of 24 species of Phyllostachys.The total content of the four flavone C-glycosides were in the range of 286.14~3838.79 mg/kg.The maximum content was found in P.nigra,and its content was 3838.79 mg/kg.The total concentrations of four flavone C-glycosides in P.propinqua were in the range of 1371.15~4367.01mg/kg in the 4 months,and the maximum content was found in January,and its content was 4367.01mg/kg.Flavonoids were abundant in bamboo leaves of 24 species of Phyllostachys.The method had good reproducibility and high accuracy,and provides a basis for quality control of flavone C-glycosides of bamboo leaves.In summary,this study established a qualitative and quantitative method for flavonoids in bamboo leaves.The differences of the content of main flavonoids in 24 bamboo leaves were compared,which provided a research basis for the utilization of bamboo leaf flavonoids.
