In recent years,cancer has become one of the major diseases that threaten human life and health,second only to cardiovascular and cerebrovascular diseases.Traditional cancer treatment methods such as chemotherapy and radiation therapy are toxic to cancer patients and may cause complications.Therefore,finding new treatment methods and developing efficient and low-toxic drugs to prevent and treat cancer are the current research hotspots of domestic and foreign communities of medicines and drugs.In recent decades,with the development of biology,microbiology and genetics,a variety of natural products with antitumor activity have been discovered and mass-produced for clinical use.Our research group has consulted a large number of literatures and experimental screening in the previous period,and found that the Chinese herbal medicine Cardamom contains a variety of active ingredients,including antitumor active ingredients.Therefore,our purpose is to explore a set of steps to isolate and purify the active ingredients in Chinese herbal medicine cardamom,and to study the biological activity of the active ingredients obtained by isolation and purification.We used alcohol precipitation,vacuum normal phase column chromatography(VLC column),C18 reversed phase column chromatography(ODS column),gel column hierarchy,thin layer chromatography and preparation of liquid phase to separate from Chinese herbal medicine cardamom.Seven compounds were isolated and purified,i.e.,Alpinetin,Pinocembrin,and Trans-3,5-dihydroxy-1,7-diphenyl-hept-l-ene,Cardamonin,Vanillic acid,P-counmaric acid,Biflorin.Among the compounds,P-coumaric acid and Biflorin are anti-cancer active ingredients that isolated and purified from cardamom for the first time.MTT colorimetric method was used to detect the effects of P-coumaric acid and Biflorin on human breast cancer MCF-7 cells,human liver cancer Bel-7402 cells,human leukemia K562 cells,and human normal cell breast MCF-10 A cells.Value-added inhibition rate.The experimental data were collated to obtain the half inhibitory concentrations of P-coumaric acid on human breast cancer MCF-7 cells and human liver cancer Bel-7402 cells: 0.0146 mg/ml and 0.0169 mg/ml,respectively.The half inhibitory concentrations of cancer MCF-7 cells,human liver cancer Bel-7402 cells,and human leukemia K562 cells were: 0.1076 mg/ml,0.3285 mg/ml,and 0.2270 mg/mg.Experiments have shown that P-coumaric acid has good antitumor activity against other tumor cells except leukemia K562 cells,and has no inhibitory effect on human normal breast MCF-10 A cells;Biflorin has good broad-spectrum antitumor The activity is the strongest inhibitory effect on human breast cancer MCF-7 cell line,and has no inhibitory effect on human normal breast MCF-10 A cell line.This study isolated and purified P-coumaric acid and Biflorin from the Chinese herbal medicine cardamom,and their activities in vitro antitumor activity were studied,laying a foundation for its subsequent antitumor mechanism research and drug production;similar components for traditional Chinese medicine.The extraction provides a technical basis;this study also provides a reference for the development of antitumor drugs and the prevention of malignant tumors,which is of great significance.
