Objective:To prepare and measure the size and Z electric potential of the mixed micelle,which is synthesized by plurnoic-doxorubicin/ plurnoic-alendronate.Methods:The anti-bone tumor drug doxorubicin(DOX)and amphiphilic block copolymer Pluronic123(P123)were chemical bonded to prepare the conjugate P123-DOX.Then the bone targeted drug Alendronic(ALD)and amphiphilic block copolymer P123 compounded the conjugate of P123-ALD.And the structures of the two conjugates were characterized by 1H-NMR.The new conjugates prepared a mixed micelle P123-DOX/P123-ALD,and the particle size and Zeta potential of mixed micelle were measured by particle size and surface potential analyzer.Results:Through the observation of 1H-NMR spectra of P123-DOX and P123-ALD,it was found than the peaks generated by amide bond in P123-DOX and p123-ALD were located at 8.1-8.2 and 7.7-7.8 respectively.The particle size and Zeta potential of P123-DOX,P123-ALD and P123-DOX/P123-ALD were measured by particle size and surface potentiometer.The results showed that the particle size of P123-DOX,P123-ALD and P123-DOX/P123-ALD were(103.7±2.56)?(123.4±1.91)?(71.3±0.10)nm respectively.All met the requirements of micelle size.The Zeta potential of P123-DOX,P123-ALD and P123-DOX/P123-ALD were(-29.4±0.67)?(-16.0±0.98)?(-17.3±1.1)mv respectively.Conclusion:P123-DOX and P123-ALD are successfully synthesized by chemical bond coupling of doxorubicin and alendronate with Pluronic123,respectively.P123-DOX and P123-ALD synthesize the mixed micelle P123-DOX/P123-ALD.The prepared mixed micelle is nanometer size and may be used as a new tumor delivery system.
