Allergy is an abnormal immune response of the body,usually accompanied by inflammation or tissue damage.Type I allergy is a common clinical allergic disease,and its related diseases or pathological processes can be local or systemic.Allergic diseases are characterized by many causes,long course and easy recurrence.The current dosage forms and treatment methods can not effectively improve the compliance of patients.Orodispersible films are promising drug delivery systems for customized medicines as it provide an alternative approach to increase consumer acceptance by advantages of rapid dissolution and administration without water.3D printing is a rapid prototyping technology,which creates solid objects layer by layer through3 D models obtained by computer aided design or 3D scanning.It is is an effective way to get instant formula and personalized drugs on demand by3 D printing.In this paper,the semi-solid micro extrusion 3D printing technology is applied to precisely print the personalized levocetirizine hydrochloride orodispersible films.The aim of this study was to develop a platform to support the realization of tailored treatments suitable for the extemporaneous production of ODFs by semi-solid extrusion 3D printingto improve the compliance of patients with medication and improve the safety of drug treatment.Orodispersible films of levocetirizine hydrochloride with personalized dosage was designed through the study of the usage and dosage of levocetirizine hydrochloride before the study.The appropriate film-forming materials and printing slurry concentrations were studied.The optimal formulation of levocetirizine hydrochloride oral orodispersible films was hydroxypropyl methylcellulose E15: levocetirizine hydrochloride:pregelatinized starch: maltitol: sucralose at a ratio of 64:10:10:15:1 by study the types and dosage of fillers,flavoring agents and plasticizers.Through the study of the process and key printing parameters in the printing process,the appropriate process parameters were explored.According to the relationship between the volume of the theoretical model and the content of the drug,orodispersible films in dose of 1.25 mg,2.5mg and 5mg of levocetirizine hydrochloride was designed and prepared.The3 D printed orodispersible films own smooth appearance,good mechanical properties,dissolution,contact angle and dose accuracy.The drug content and content uniformity meet the requirements of Chinese Pharmacopoeia.3D printing semi-solid micro extrusion technology can be used as a new small dose personalized orodispersible films preparation method.At the same time,this study provides a new method for the preparation of extemporaneous personalized inorodispersible films,and provides areference for the application of 3D printing orodispersible films in hospital or pharmacy and other personalized medical fields.
