MARVELD1 was discovered in previous research in our laboratory.It is located on the human chromosome 10q24.2,it has two extons,its full-length m RNA is 3238 bp,encoding a 18.9 k Da protein.MARVELD1 has important biological functions in cell proliferation,DNA damage repair,metastasis and invasion,etc.but at present,little is known about the research on apoptosis.Previous studies found that MARVELD1 can reduce the level of oxidative stress in cells under cellular stress.In addition,it was also found that MARVELD1 can inhibit the apoptosis in liver cancer cells induced by arsenic trioside by enhacing the expression of TRXR1.Therefore,the study preliminary discussed the function and molecular mechanism of MARVELD1 in the apoptosis pathway induced by X-ray irradiation in this paper.The study detected cell viability,the levels of intracellular reactive oxygen species and apoptosis levels,and found that X-ray radiatio can induce apoptosis in He La cells and MEF cells.After 2 Gy,4 Gy and 8 Gy of X-ray irradiation,the overexpression of MARVELD1 in He La cells can significantly increase the cell viability,decrease the level of intracellular reactive oxygen species(ROS)and reduce the level of apoptosis.In MEF cells,MARVELD1 knockout can significantly reduce the cell viability,increase the level of ROS and apoptosis.The m RNA expression of apoptosis related genes was detected by real-time quantitative PCR,and found that MARVELD1 knockout could significantly reduce the m RNA expression of FADD and Bcl-2,up-regulating the expression of bax,caspase 9 and caspase 3,and down-regulating the ratio of Bcl-2/Bax to promote apoptosis.In conclusion,the study confirmed that MARVELD1 in He La and MEF cells may reduce the apoptosis caused by X-ray irradiation by reducing the level of ROS in cells.In MEF cells,MARVELD1 effects on apoptosis induced by X-ray radiation through FADD pathway,and its knockout will promote apoptosis by down-regulating Bcl-2/Bax m RNA expression.
