Point-of-care testing(POCT),as an important part of in vitro diagnosis,exhibits advantages of rapid analysis,simple operation,easy operation and low cost,etc.It has attracted extensive attention from researchers in clinical diagnosis,environmental quality monitoring,food safety testing and other fields,and has huge potential of application in these fields.Microfluidic chip is a revolutionary technical platform that has many major breakthroughs after nearly 20 years of development,and showing huge market prospects.Portable detection device is an important research direction of the current instrument analysis.With the characteristics such as low cost and simple operation,the miniaturized analytical devices for household use have broad space of development.In this thesis,we developed portable detection devices for POCT by using microfluidic chips as the main platform and examined their application.This dissertation consists of four chapters:In the first chapter,we briefly introduced the field of POCT and its development and application status,focusing on the common materials of microfluidic chip technology,production process,channel modification methods and detection technologies.Smartphone-based detection and chemiluminescent detection in portable detection equipment were also introduced.In the second chapter,we used smartphone and 3D printed optical imaging system as photo detection devices.Microfluidic chips with 'vertical channels' array were used for the TMB colorimetry was also used to detect HIV p24 antigen by the aid of mobile phones.The detection conditions for p24 antigen were optimized,with a final detection limit of 0.022 ng/mL attained in human serum sample.In the third chapter,based on the integrated POCT system of "vertical channel" array microchip and smartphone as described in Chapter 2,the detection of the type A of botulinum toxin light chain protein was performed and the detection conditions for the type A botulinum toxin light chain protein were optimized,a detection limit of 2.8 ng/mL was obtained.In the fourth chapter,a chemiluminescent immunoassay device was constructed using 3D printing,PMT was used as the luminescent signal detector.Its detection performance wasevaluated,and a comparison with a commercial microplate reader was performed.And the detection limit for HRP of 0.26 pg/mL was achieved.
