?-Acyloxy ketone compounds and their derivatives are an important class of skeletons in organic synthesis, which demonstrate widely applications in biology, medicine, material scienece and other fields. The traditional synthetic route to?-acyloxy ketones mainly depends on the direct esterification reaction of well-performed?-hydroxy ketones. However, the substrates of the this protocol is not easily available, which makes it difficult to achieve the universal and efficient synthesis of?-acyloxyketones.Therefore,it is an urgent challenge for chemists to establish new methods to the synthesis of?-acyloxy ketone compounds.Carbon-carbon bonds are the most basic and widespread chemical bonds in organic compounds. The reactions involving the cleavage of carbon-carbon bonds have been distributed in each corner of our daily life as well as the industrial production. The first chapter of this thesis systematically summarizes the synthesis method of?-acyloxyketones and the cleavage reaction of carbon-carbon bond. The C(sp~2)-C(sp~3)bond in 1,3-dicarbonyl compounds is a special kind of carbon-carbon bond due to the high reactivity. In recent years,a lot of elegant works on carbon-carbon bond cleavage of 1,3-dicarbonyl compounds have been reported. However,in most cases, transition metals, especially for cooper catalysts were often used. This thesis disclose a transition metal-free and base-promoted reaction of1,3-dicarbonyl compounds and 3-halo-1-propanols,delivering varieties of?-acyloxy ketone compounds in good to excellent yields. The second chapter of the thesis includes the detailed optimization of reaction condition and the expansion of substrate scope. The large-scale reaction still proceeded smoothly in a high yield, suggesting its potential industrial applications.
