With the intensification of environmental pollution,people's skin barriers are more and more easily damaged,such as harmful microorganisms,chemical carcinogens,physical radiation,etc.,which in turn leads to skin barrier diseases such as dry skin syndrome and psoriasis.These skin barrier properties The disease brings huge life and mental stress to the patients.Hydroxytyrosol has good antioxidant,anti-inflammatory,antibacterial,anti-tumor and other biological activities,but due to the instability and low bioavailability caused by its polyphenol structure,lecithin inclusion technology is used to carry out Treatment is of great significance,but lecithin inclusion technology usually requires a lot of time and effort to carry out experimental exploration and verification,which is not conducive to the development of efficient scientific research.Therefore,this paper proposes to use dissipative particle dynamics(DPD simulation)to simulate the inclusion effect of lecithin on hydroxytyrosol,optimize the inclusion process,and verify the effect of the simulated inclusion compound through experiments.At the same time,the application of hydroxytyrosol and its inclusion compound in the repair of two damaged skin barriers of mouse psoriasis and mouse xeroderma,explore the effect of hydroxytyrosol and its inclusion compound on the damaged skin barrier And mechanism.In this paper,first,computer dissipative particle dynamics(DPD simulation)is used to simulate the inclusion effect of lecithin and hydroxytyrosol in different solvent systems,including water solvents,low-carbon alcohol solvents,and oil solvents.At the same time,the DPD simulated inclusion compound was verified,including preparation,transmission electron microscopy characterization,inclusion rate determination,in vitro sustained release experiment,etc.,to explore the prediction and guidance of DPD simulation on the inclusion of hydroxytyrosol.The experimental results show that the lecithin inclusion compound of hydroxytyrosol has a good inclusion effect in the water system,and the inclusion rate can reach 43.74%,which has a significant in vitro sustained release effect.Then,the hydroxytyrosol and its lecithin inclusion compound was applied to the mouse psoriasis-like animal model,and the hydroxytyrosol and its lecithin were explored through experiments such as skin PASI score,skin transdermal water loss rate and skin HE staining analysis.The effect of inclusion complex on skin repair in psoriasis mice.The results show that hydroxytyrosol and its lecithin inclusion compound have a certain relief and repair effect on skin psoriasis in mice.Through the real-time fluorescence quantitative nucleic acid amplification detection system(q-PCR)experiment on the transcription of key inflammatory factors in mouse skin psoriasis,it was found that hydroxytyrosol and its lecithin inclusion compound have a significant inhibitory effect on IL-17 inflammatory factors The low concentration of lecithin hydroxytyrosol inclusion compound has a stronger inhibitory effect on IL-17 than the hydroxytyrosol aqueous solution system.The results of enzyme-linked immunosorbent assay(ELISA)showed that hydroxytyrosol and its lecithin inclusion complex mainly down-regulated the m RNA transcription of IL-? to inhibit the occurrence of psoriasis in mice.Finally,in the mouse model experiment of acetone-ether skin dryness,it was found that hydroxytyrosol and its lecithin inclusion compound have a certain relief and repair effect on dry skin damage,which is mainly manifested in the reduction of skin cuticle and inflammation of epidermal cells The degree of infiltration is reduced.Analysis of protein genes of dry and damaged skin by q-PCR.Hydroxytyrosol and its lecithin inclusion compound have a certain regulatory effect on the expression of KRT10 m RNA,KRT1 m RNA,FLG m RNA,and IVL m RNA in the skin epidermis.Lecithin The hydroxytyrosol inclusion compound is significantly stronger than the hydroxytyrosol aqueous solution in regulating effect.In summary,we have demonstrated that computer simulation-assisted inclusion technology is instructive in the hydroxytyrosol lecithin inclusion process and that hydroxytyrosol and its lecithin inclusion compounds have a certain relief and repair effect on the skin barrier.The research results provide a certain research basis for hydroxytyrosol and its lecithin inclusion complex in skin barrier repair and related disease treatment.
