The incidence of cancer has increased sharply every year,and it has always been a difficult problem for humans to overcome.How to improve the drug efficacy,facilitate operation,and reduce toxic and side effects in cancer treatment is a hotspot of scholars from various countries.Microspheres are a good drug delivery system that can be delivered to patients by oral,intravenous,subcutaneous injection,or transcatheter arterial chemoembolization(TACE).Drug-loaded embolization microspheres are a new type of pharmaceutical preparation that uses TACE to continuously and slowly release chemotherapy drugs at the tumor site to increase the drug concentration and reduce adverse reactions.At the same time,embolize the blood vessels of the tumor tissue,interrupt the blood supply,prevent the tumor cells from drawing nutrients,and inhibit Its growth achieves the purpose of treating cancer,and has important research significance especially for advanced liver cancer.In order to achieve this goal,pectin/polyvinyl alcoholcontaining embolization microspheres were prepared using natural polysaccharide gum and polyvinyl alcohol as basic carriers,and their anti-tumor activity was preliminary evaluated.The specific research contents and conclusions are as follows:(1)The emulsification-chemical cross-linking method was used to study the preparation process of pectin/polyvinyl alcohol microspheres,and the best conditions for preparing pectin/polyvinyl alcohol microspheres were obtained: 3.7 g emulsifier SP-80 mixed liquid paraffin Oil phase,pectin: polyvinyl alcohol = 7: 3 The mixed solution is the water phase,according to the oil-water volume ratio of 10: 1,stir to form a water-in-oil system,and then add the cross-linking agent glutaraldehyde 0.29 m L at 40 ? Under stirring for 3 h,microspheres with different particle sizes ranging from 1-1000 ?m were prepared by controlling the stirring rate.(2)Using doxorubicin(DOX)as a model drug,drug-loaded pectin/polyvinyl alcohol microspheres were prepared and their properties were studied.The maximum drug loading of pectin/polyvinyl alcohol microspheres was 20.0%;the drug release performance was studied,and the cumulative drug release rate reached 80.7% at 72 h;the microsphere swelling rate was 202%;the microspheres were spherical after drug loading and drug release Regular,complete structure and smooth surface;cytotoxicity experiments showed that the microspheres have good antitumor activity.(3)Further,magnetic pectin/polyvinyl alcohol drug embolization microspheres were prepared and their properties were studied.The microspheres are spherical,uniformly distributed,have superparamagnetism,and have a saturation magnetization of 6.2 emu/g,which can meet the magnetic strength requirements in biomedicine and lay the foundation for the magnetic control of drug-loaded embolization microspheres.
