With the rapid development of our country's breeding industry,the problem of environmental pollution caused by livestock and poultry manure is becoming more and more serious,the anaerobic fermentation method has attracted much attention as one of the important means for treating livestock and poultry manure.Anaerobic fermentation is the main method for processing solid wastes such as buffalo manure,natural anaerobic fermentation has the problems of long maturation time,large nutrient loss and large secondary pollution.The addition of exogenous bacterial agents in anaerobic fermentation has been widely confirmed to enhance the activity of microorganisms to decompose organic matter,accelerate the decay process,and improve the quality of anaerobic fermentation products.This experiment is aimed at the current situation of less beneficial indigenous microorganisms in traditional anaerobic fermentation in our country,which leads to slow fermentation and long maturing time,by comparing the harmless effects of microbial inoculants A and B applied to the anaerobic fermentation of buffalo manure alone and mixed,it laid the foundation for the production of mattress pads and the establishment of an integrated feeding system for bed-barnlot integration.The following is the main research content and results of this paper:1.In this experiment,a total of 4 anaerobic fermentation systems for buffalo manure were added,with the addition of self-researched bacteria A,the addition of self-researched bacteria B,the simultaneous addition of A and B self-researched bacteria,and no addition of bacteria,by measuring the five indicators of temperature,p H,water content,carbon to nitrogen ratio(C/N)and seed germination index(GI)during the anaerobic fermentation process,to determine the degree of maturity of buffalo manure anaerobic fermentation.The result is as follows:The temperature of the three experimental groups with added microbial inoculum during the anaerobic fermentation heating and high temperature periods was significantly higher than that of the blank group without added inoculum(P<0.05),its temperature rises more quickly,and the residence time in the high temperature period is longer.This shows that the addition of this self-developed bacterial agent during the anaerobic fermentation of buffalo manure can effectively increase the reaction temperature of anaerobic fermentation,at the same time increase the duration of the high temperature period of anaerobic fermentation and improve the efficiency of anaerobic fermentation.Comparing the index data of the three experimental groups with added bacterial agents in Group A,Group B and Group A+B,we can find that the overall change trend of temperature,p H,water content,carbon to nitrogen ratio(C/N),and seed germination rate(GI)of each group were roughly the same.However,the range and rate of change of each index in the A+B group were the highest during the high temperature period,and the maximum temperature of the A+B group reached 53.4°C and stayed above 50°C for more than 5 days,meanwhile,during the anaerobic fermentation process,the p H value changed between 7.5 and 9.0,the water content decreased from 59.43% to 35.65%,the carbon-to-nitrogen ratio(C/N)at the end of the anaerobic fermentation(C/N)<20,and the seed germination rate(GI)> 80%,all of the above have reached various evaluation indexes of anaerobic fermentation.While the other two experimental groups did not meet some of the conditions.Therefore,it can be concluded that the anaerobic fermentation effect of adding A and B bacteria simultaneously during anaerobic fermentation is better than adding A or B bacteria alone.2.In this experiment,the mixed bacteria group(A+B)anaerobic fermentation initial sample(Z1),high temperature stage sample(Z2,Z3),end stage sample(Z4)were used to identify strains to investigate and compare the changes of bacteria in each period.The analysis found that:A total of 5 strains were cultivated in the sample Z1 in the initial stage of anaerobic fermentation.10 strains were cultivated from 2 samples Z1 and Z2 in high temperature period.Five strains were isolated and cultured from sample Z4 at the end of anaerobic fermentation.Most of the bacterial species isolated and cultured in the samples at high temperature during anaerobic fermentation are mesophilic bacteria(Bacillus subtilis,high temperature actinomycetes,etc.)with strong ability to decompose organic matter.Comparing the types of bacteria isolated and cultured from the samples at the beginning of the anaerobic fermentation and the samples at the end of the anaerobic fermentation,we can find the pathogenic and harmful bacteria(E.coli,Clostridium,etc.)in the pile after the anaerobic fermentation is completed)has decreased or disappeared.In summary,this experiment successfully established an anaerobic fermentation system for buffalo manure.The treatment of buffalo manure has basically achieved the purpose of reducing and recycling the waste of the farm.At the same time,the identification of bacteria in different periods of anaerobic fermentation also shows that the addition of self-researched bacterial agents A and B in the anaerobic fermentation process has a significant effect on the harmless treatment of breeding waste c buffalo manure.
