Synthesis Of Chitosan-based Hydrogel Based On Supramolecular Assembly

Fullerene(C₆₀)is one of the most unique natural molecules.C₆₀ has photosensitizing activity,and can be used for photodynamic therapy while being administered in vivo.These advantages make it an excellent drug carrier,which is expected to achieve targeted positioning and sustained and controlled release of drugs in the body.This article creatively developed a new method for the synthesis of fullerol.After the electrophilic addition reaction of fullerol(C₆₀)and liquid bromine?Br₂?under dark conditions produces bromofullerene(C₆₀Br₂₄),then in a NaOH alkaline solution environment,C₆₀Br₂₄ and ethanol undergo nucleophilic substitution After the reaction,a new type of fullerol containing both ethoxy?-OCH₂CH₃?and hydroxyl?-OH?in its structure was obtained,which was named ethoxyfullerol.Thin layer chromatography?TLC?-column chromatography-thin layer chromatography to purify the product,and characterize and analyze it by FT-IR,¹H-NMR,elemental analysis and TG/DTG,and its chemical formula of this new fullerol was finally determined as C₆₀?OCH₂CH₃?₈?OH?₁₆.In biomedicine and other fields,the new ethoxyfullerol(C₆₀?OCH₂CH₃?₈?OH?₁₆)improves the inherent defect of C₆₀ being completely insoluble in water.The solubility test shows that it is a kind of C₆₀ derivatives.The crystal culture results show that the polarized microscope has general crystal characteristics,but does not obviously have the characteristics of single crystals.Some methods of modifying chitosan were explored,and derivatives of chitosan-g-?-cyclodextrin?CS-CD?were prepared.Based on the theory of supramolecular chemistry,the CS-CD and ethoxyfullerol physically interact with the hydroxyl groups carried by them to carry out supramolecular assembly.This method of using drug molecules as the cross-linking point of the drug controlled release system provides a new solution and basis for the intelligent release of drugs.
