Due to the unique surface effect,superparamagnetism and tunneling effect of magnetic nanoparticles,they can be used as a carrier for targeted treatment of cancer drugs.At present,the traditional methods of cancer treatment are mainly co-cultured with drug carriers.Because of the shortcomings of not being able to process specific targeted cells and can't be able to grasp changes in target structure in real time,this method is limited to a certain extent in the process of cancer diagnosis and treatment research.Therefore,a new method is needed to achieve targeted treatment of cancer drugs.In this thesis,magnetic nanoparticles are delivered to specific cells by magnetic micromanipulation system to achieve real-time localization of cancer cell targets and observe and analyze the micro-nano structure changes of targeted cells.1.Aiming at the problem of difference in movement or pickup in the research of manipulating magnetic nanoparticles,the variation law of surface roughness and adhesion force of mica substrate was obtained,through the modification of mica substrate by 3-aminopropyltriethoxysilane(APTES).Based on the theoretical analysis of the ABAQUS simulation model,the mechanical simulation model of magnetic probe picking up magnetic nanoparticles was established,and the magnetic nanoparticles were manipulated by magnetic force microscope.The mechanical characteristics of magnetic nanoparticles during the manipulation were compared and analyzed through the theoretical and experimental results.Mica substrates were modified by different concentrations of APTES solutions,and the influence of substrates with different roughness on adhesion during the manipulation were analyzed.2.Aiming at the shortcomings in the process of targeted implantation by co-culture,a simple targeted implantation method was proposed.This method can obtain the structural changes of targeted cells during the targeted implantation.Based on the principle of micromanipulation technology,the mechanical simulation model of probe penetration was established,and the stress changes of cells during the targeting process were analyzed.Combined with the experiments,the magnetic nanoparticles targeting implanted cells with fluorescent marker,and observation whether particles into target cells by fluorescence microscope.Changes in surface morphology,Young's modulus,and adhesion of cancer cells before and after targeted implantation were observed by atomic force microscopy.The effects of targeted implantation on the cells were analyzed and summarized.The experimental results show that this method can accurately target magnetic nanoparticles into the cell,and provide theoretical basis and experimental means for the research of magnetic nano-drugs and cancer diagnosis and treatment.
