Ganoderma lucidum,as an important resource of national medicine and food homology,is one of the "top ten Wanyao drugs".It is used as a health promoter because of its anti-tumor and immunomodulatory effects.In this paper,Ganoderma lucidum was used as material to investigate the apoptosis mechanism of He La cells induced by Ganoderma acid A and the antitumor effect of Ganoderma acid A on mice.Finally,the regulation of Ganoderma acid A on He La cells was determined from m RNA level by using transcriptome technology.The results of this study clarify the anticancer mechanism of Ganoderma acid A,and establish the basis for the development of ganoderma as a special medical food.Ganoderma acid A induces He La cell apoptosis and its mechanism.When He La cells were treated with standard Ganoderma acid A,the proliferation of He La cells was significantly inhibited by Ganoderma acid A,with IC50 of 39.8 ?m in 24 hours.Ganoderma acid a blocked cell cycle in G2/M phase and induced apoptosis.After Ganoderma acid a treatment,the expression of genes related to cell cycle changed,the expression of CHK2,CDK1,cdc2 and cyclin B decreased,and the expression of cdc25,p53 and p21 increased.The expression of Bax,Bak,BIM,caspase-9,Caspase-3,TNF-R1,Caspase-8,p21,puma,cytc,p-Akt,PARP,Apaf-1,p38 AMPK and Akt,Bcl-2 and Bcl XL in the above pathway were significantly increased after treatment with Ganoderma A.Therefore,Ganoderma acid a can induce He La cell apoptosis by regulating the expression of related genes in death receptor signaling pathway and mitochondrial signaling pathway.The effect of Ganoderma acid A on the m RNA level of He La cells.According to the IC50 value of the previous MTT experiment,He La cells were treated with Ganoderma acid A at a concentration of 40?M.After treatment with Ganoderma acid A for 24 hours,there were 4511 up-regulated genes and 3875 down regulated genes in He La cells.After the treatment of Ganoderma acid A,the differential genes were mainly enriched in p38 MAPK,p53 and PI3 K signaling pathways.
