As a new type of edible fungus,Auricularia cornea var.Li(ACVL)is still in the promotion stage in the market.In recent years,there has been more research on its cultivation and breeding conditions,but less on its composition and biological activity.In nowadays,most people eat too fine in terms of diet,and the daily intake of sugar and fat far exceeds the content required by the body,which will inevitably lead to an increase in the number of obese people.In order to allow people to understand the value of ACVL more deeply,this experiment studied the anti-diabetic activity of its polysaccharide and developed the replacement food for obese and overweight people,which is of great significance for the future development of the activity of ACVL and its functional food.The specific research content and results are as follows:1.In this experiment,the ultrasonic assisted extraction was used to extract crude polysaccharides from ACVL,and after the protein was removed with the Sevage method,the purified the polysaccharide was separated with a DEAE-cellulose-52 column,the recovery rate is 487.23 ± 12.23 mg / g,and the polysaccharides was scanned in the ultraviolet spectrum at a wavelength of 190-400 nm.The results showed that there was no absorption peak at 260 nm and 280 nm in the polysaccharides,indicating that the polysaccharides did not contain nucleic acid and protein.The structure of the polysaccharides was determined using Fourier infrared spectroscopy.The results showed that the polysaccharide had the characteristic absorption peak of polysaccharide,and it was found that the polysaccharide had two glycosidic bonds in ? and ? configurations.2.This experiment explored the anti-diabetic activity of polysaccharides from ACVL in mice: ? Effect on blood sugar: The intragastric treatment showed that the polysaccharides can reduce blood sugar level fast in diabetic mice.? Effects on lipid metabolism: The treatment on type ? diabetic mouse model with the polysaccharides showed that it can reduce the content of triglyceride,total cholesterol,and low-density lipoprotein cholesterol in the serum of model mice and increase High-density lipoprotein cholesterol content.? The effect on SOD value,hepatic glycogen and insulin secretion in mouse serum: the polysaccharide can increase the level of SOD in mouse serum,indicating that ACVL polysaccharide can inhibit the oxidative stress.The finding,through the upregulation of liver glycogen and insulin levels,shows that the polysaccharides play an important role in the regulation of glucose metabolism in mice.?Through the observation of HE slices,it suggested that the polysaccharides have alleviated the inflammation of the kidney and liver.These results suggested that the polysaccharides of ACVL improve the regulation of lipid metabolism and insulin secretion significantly in mice.It can reduce blood sugar and body weight in mice and has obvious anti-diabetic activity.3.In this experiment,the formula of low-fat replacement powder with ACVL was studied and optimized through response surface test.The best formula was determined: the amount of ACVL powder is 21.55 %,the amount of arabinose is 7.72 %,and the amount of konjac fine powder is 8.34%,and the amount of skimmed milk powder is 15.5 %.Its dissolving quality improved and the basic powder particle was determined to be 120-140 mesh,the optimal preparing concentration ratio was determined to be 1: 6(g / mL),and the preparing temperature was determined to be 80 ?.The nutrient composition was analyzed on the low-fat powder of ACVL meal replacement and the results showed that it was in accordance with the dietary level of obese,high blood fat,and diabetic people.
