Peroxynitrite(ONOO~-)is a kind of active oxygen species which was formed by superoxide anion free radicals and nitric oxide free radicals in cells,and ONOO~-is an important kind of signaling molecule in many signaling pathways in the body.However,it could also cause irreversible damage to some biological targets such as nucleic acids,sugars,proteins,lipids,DNA and so on,which caused the generation of dysfunction in molecules and cells.ONOO~-is considered to be a key targets of many diseases including inflammation,cancer and neurodegenerative diseases.Therefore,it is of great significance for the development of novel fluorescent probes in real-time monitoring the diagnosis and treatment of clinical disease by imaging ONOO~-.In this article,three probes were synthesized for the detection of ONOO~-.1. Based on the strategy of nucleophilic reaction of carbon-carbon double bond toward with ONOO~-,a“Turn-on”fluorescent probe(DDM-Me)was designed and synthesized.The probe was successfully applied in detecting of ONOO~-.After the probe reacts with ONOO~-,there is a strong fluorescence intensity at 402 nm.The probe has advantage of high sensitivity for the detection of ONOO~-.At the same time,the probe showed good selectivity among many interfering substances.The probe also has a good response under physiological conditions.These results would provide support for the biological detection of ONOO~-in future.2. Based on the previous research,methyl group was replaced with a methoxy group to study the effect of the substituent on the detection effect of the probe.After introducing methoxy,a novel fluorescent probe(DDM-OMe)was synthesized which has a longer emission wavelength.At the same time,the probe reacts with ONOO~-in a shorter time.The probe could also detect ONOO~-sensitively.In the presence of multiple interferences,it can specifically react with ONOO~-,which revealed that the probe also have advantage of high selectivity.All in all,the spectroscopic properties of this probe meet the requirements for detecting ONOO~-,and could also provide some help of biological detection in future.3. Based on the previous research,it is known that the properties of the probe could be changed by introducing the different substituents in the structure of probe. the strong electron-donating substituent(hydroxyl group)was chosed to modify the structure of probe.Compared with the previous probe,DDM-OH has a greatly longer emission wavelength.And,the probe has high sensitivity and specificity in detecting ONOO~-.The probe could be rapidly reacted with ONOO~-.All results reveals that the probe is conducive to its detection and application in vivo.
