Traditional Chinese medicine(TCM)contained a large number of extremely complex components.In order to further apply it in the treatment of diseases and other aspects,the efficient extraction methods should be established to extract the active compounds and evaluate their quality objectively and reliably.In this study,the in-situ reaction and vesicle assisted new extraction methods of TCM were established,which could meet the requirements of economic and environmental protection and obtain satisfactory extraction effect and sensitivity.The main research contents are as follows:1. An in situ chemical reaction-based matrix solid-phase dispersion method was developed for the extraction of the hydrophilic and hydrophobic compounds in the plant Scutellariae radix.The method revealed good linearity(r~2?0.9978)for the target analytes,with the limits of detection from 12.52 to 59.76?g/kg,the limits of quantification from 41.72 to 199.20?g/kg and the method quantification limits from0.124 to 0.292?g/kg.Moreover,the recovery values of the tested compounds ranged from 86.0%to 104%.The developed method simplified the pretreatment of extraction,reduced the use of samples and dispersants,and used pure water instead of organic solvents.2. An effective method based on the matrix solid-phase dispersion microextraction was developed for the extraction of alkaloid compounds in Fritillariae Thunbergii Bulbus.Target analytes were analyzed by capillary electrophoresis coupled with quadrupole time-of-flight mass spectrometry.The results demonstrated that the investigated method had low limits of detection(1.32-1.59 ng/m L),good recoveries(86.63-98.12%),and reproducibility(relative standard deviations of peak areas<0.87%).3. A simple and effective vesicle based ultrasonic-assisted extraction method was developed for extraction of active compounds in Panax notoginseng.The target analytes were determined by ultra-high performance liquid chromatography with ultraviolet detector.Surfactant vesicle was adopted as extraction solvent.The proposed method provided good linearity in the linear range of 10-1000?g/m L with regression coefficients larger than 0.999,low limits of detection of 27.64-55.67ng/m L,good precision with relative standard deviations below 0.35%,and satisfactory recoveries of 83.84-90.92%for tested saponins.4. A simple and sensitive in-suit antioxidation assisted with matrix solid phase dispersion method for the extraction and separation of chiral flavonoids(narirutin,naringin,hesperidin,neohesperidin)in various citrus plants was established using ion mobility quadrupole time-of-flight high resolution mass spectrometry.The developed antioxidant technology was a green method that used minimal samples and organic solvents.The chiral separation method used a conventional chromatographic column without chiral additives and could quickly separate flavonoids,thus was considered as a green method.The two methods was fast,which greatly saved the use of organic solvents.In this study,collision cross sections values were beneficial to obtain the structure information of target compounds.According to the experimental results,the method showed satisfactory LODs(3.70-6.52 ng/m L)and good selectivity and recoveries(96.78-104.67%)for the four flavonoids in citrus plants.
