Chronic infectious wounds are a serious problem in the world's wound diseases,which have a serious impact on people's lives.Infectious wounds make the wound heal more slowly because of the bacteria,and antibiotics can effectively treat microbial infections.However,because of the abuse of antibiotics in recent years,excessive bacteria have developed drug resistance,which has led to serious problems in the treatment of bacterial infection wounds.Therefore,the development of a new type of new dressing containing antibacterial effect has become an urgent problem to be solved.In this paper,p-toluenesulfonyl chloride and diols(triethylene glycol,triethylene glycol monobutyl ether,diethylene glycol monobutyl ether)compounds can be synthesized to synthesize p-toluenesulfonate compounds,and then pass SN2 pro The nuclear substitution reacts with the positively charged triphenylphosphine to synthesize a novel quaternary phosphonium salt antibacterial material.The polyamine hydrochloride was grafted onto the sodium alginate chain by a crosslinking agent,and then formed into a hydrogel by a freeze-thaw method with polyvinyl alcohol.Physical and chemical properties of antimicrobial materials and hydrogels were characterized by a series of characterization methods.Finally,the quaternary phosphonium salt was combined with hydrogel and tested for antibacterial properties and in vitro cytotoxicity.The research results are as follows:(1)Successfully synthesized three kinds of new quaternary phosphonium salt antibacterial materials(triethylene glycol,triethylene glycol monobutyl ether,diethylene glycol monobutyl ether).The synthesis process is simple,the conditions are easy to realize,and the purification steps after the reaction are simple.High yield;successfully prepared sodium alginate-dopamine/polyvinyl alcohol hydrogel.The hydrogel preparation method has mild reaction conditions and simple operation steps,and the prepared hydrogel has strong physical and mechanical properties and good water absorption performance.(2)Three kinds of quaternary phosphonium salt antibacterial materials have significant antibacterial effects against Gram-negative bacteria-E.coil and Grampositive bacteria-S.aureus,among which three ethylene two Alcohol-bistriphenyl-ptoluenesulfonate has the strongest antibacterial property due to the combination of triphenylphosphine on both sides;the composite antibacterial hydrogel also has significant antibacterial effect on E.coil and S.aureus,showing a broad spectrum.Antibacterial.(3)The three new quaternary phosphonium salts are not toxic at a concentration of 2.0 mg/mL or less,and show mild toxicity to mouse 3T3 fibroblasts at high concentrations,and their biocompatibility is good;Antibacterial hydrogel has no toxicity or minimal toxicity to mouse 3T3 fibroblasts.The combination of quaternary phosphonium salt material and hydrogel greatly reduces the toxicity of quaternary phosphonium salt,thus acting as antibacterial and non-toxic to cells.(4)Experimental results of mouse full-thickness skin lesions.The experimental results show that the new antibacterial hydrogel can significantly promote the healing of infected wounds and accelerate the repair of wounds.The composite antibacterial hydrogel prepared in this study has strong physical and mechanical properties,good biocompatibility and can accelerate wound healing,has significant antibacterial effect,and has a good application prospect in antibacterial dressings.
