Photothermal therapy is a minimally invasive treatment for cancer.In order to reduce the side effects of photothermal therapy and improve the efficiency of treatment,CT imaging can be used to confirm the location of tumors and real-time monitor the photothermal treatment process.At the same time,the toxicity of nanomaterials is closely related to the inflammation reaction in the tumor area.Liver and spleen prevent nanomaterials from reaching the target site.Macrophages deliver nanoparticles which are not easily recognized by the body,improve the stability of nanoparticles and reduce the toxic side effects of nanomaterials.The main contents of this paper are as follows:(1)A kind of composite nanoparticles was prepared,which integrates X-ray computed tomography imaging and photothermal therapy.A new type of photocatalyst,BiOI,was used as CT contrast agent and wrapped in hollow copper sulfide(CuS)nanocage.The size,morphology and absorption spectra of BiOI@CuS nanoparticles were characterized by DLS,TEM and ultraviolet-visible-near infrared absorption spectroscopy.The particle size of BiOI@CuS nanoparticles was 150 nm.After 5 min of laser irradiation,the temperature of BiOI@CuS solution with a concentration of 1.00 mM was increased by 23.8?,and the circulation was good.The chemotherapeutic drug adriamycin was loaded on nanoparticles.Experiments in vivo and in vitro showed that the composite nanoparticles had excellent tumor killing effect.After 3 min of irradiation with 980 nm laser for 14 days,the tumor volume of mice was reduced by about 50% and the CT imaging effect was excellent.(2)Furthermore,on the basis of photothermal therapy combined with chemotherapy,aspirin was added to reduce the inflammation of tumor tissue.The incubation time of macrophages loaded with BiOI@CuS nanoparticles was the most effective for 2 h.The toxicity of nanoparticles to RAW264.7 cells was less.After loaded with BiOI@CuS nanoparticles,RAW264.7 cells tended to have stronger killing effect on tumor cells.RAW264.7 cells transfected with BiOI@CuS nanoparticles have stronger invasion and migration ability in the environment of cancer cells,which is helpful to enhance the targeting ability of the system to cancer tissues.In this paper,to combine chemotherapeutic photothermal therapy with CT imaging organically,we combine CT contrast agent and photothermal materials,loaded chemotherapeutic drugs.We reduce inflammation of tumor tissue by anti-inflammatory drugs,and use macrophages to deliver nanoparticles.This kind of nanoparticles have outstanding CT imaging effect and good photothermal treatment effect.The high-security integrated diagnostic and therapeutic nanoparticles have good application prospect in cancer diagnosis and treatment.
