Protein based hydrogels can be used in many fields like tissue engineering,drug release,regenerative medicine,etc since they have good biocompatibility/degradability and low cytotoxicity/immunogenicity.However,poor mechanical properties limit the development and application of protein hydrogels,and current methodsare usually dedicated,lack of effective energy dissipation mechanism,and single source.The large amount of reactive functional groups of bovine serum albumin?BSA?made its easily build hydrogel through multiple ways.Thus,in this thesis,BSA was used as a protein template to prepare three kinds of high-strength protein hydrogels,and the performance of each hydrogel system was systematically investigated.The main contents are as follows:?1?Dual-crosslinked BSA gel was prepared through the coupling reaction of amino group with carboxyl group via EDC/NHS and thermal denaturation of protein.Under optimal condition,the hydrogel showed excellent compressive properties(Ec of 971 kPa,?c,f of 89%,?c,f of 115 MPa),self-recovery property?the recovery rate of stiffness/toughness was 85%/91%when hydrogel recovered 10 min at room temperature,respectively?and fatigue resistance(no obvious change in appearance after 200 times compression with?max of 50%).?2?Hybrid crosslinked BSA/PAAm DN gel was prepared through thermal denaturation of protein and double-network concept.Under optimal condition,the hydrogel showed excellent mechanical properties(Et of 795.5 kPa,?t,f of 47.30 mm/mm,?t,f of 0.48 MPa,W of 14.88 MJ/m3,?c,f of 50 MPa)and self-recovery property?recover at 80?for 30 min,stiffness recovery of 94%and toughness recovery of 80%?.In addition,the hydrogel showed strong surface adhesion to various nonporous solid surfaces?interfacial toughness of 1176-2827 J/m2?.?3?Dynamic covalent bond/Covalent bond hybrid crosslinked THPC-BSA/PAAm DN gel was prepared through the reversible reaction of THPC with amino groups and double-network concept.Under optimal condition,the hydrogel showed excellent tensile properties(?t,f of 0.67 MPa,?t,f of 34.17 mm/mm,Et of 136 kPa,W of 14.68 MJ/m3)and extremely fast self-recovery property?recover at room temperature for 7 min,stiffness recovery rate is 90%,toughness recovery rate is 99%?.
