Food-borne pathogens are one of the main causes of food safety problems,which have been threatening the health of consumers.Salmonella can not only cause human and animal infections,but also cause human poisoning by contaminating animal-derived food.Salmonella is one of the most common food-borne pathogens,and is the most important food safety microbiological detection.With the progress of science,many new detection methods have been developed.The traditional method of Salmonella detection is complicated,the culture medium configuration time is long,and the detection cycle is long,so it can not meet the needs of rapid detection in food industry.In this paper,the formula components of the pre-prepared culture medium for Salmonella test pieces are screened,and then the pre-prepared culture medium is attached to a culture medium carrier consisting of a bottom plate,cold gel,an air permeable upper membrane and the like to form a Salmonella rapid detection test piece so as to improve the application value of the product.In this paper,the components of the formula of the pre-prepared culture medium for the rapid detection of Salmonella are mainly screened.Firstly,single factor experiments are carried out to determine the optimal concentration of each component,and then orthogonal experiments are carried out to optimize the formula.The final formula of the rapid detection of Salmonella is: peptone 10g/L,yeast extract 3.5g/L,sodium chloride 5g/L,glucose 4.5g/L,calcium carbonate 2mg/L,sodium pyruvate 2.5g/L,magnesium sulfate 4.5mg/L,nicotinamide 0.2g/L,calcium carbonate 3g/L,neomycin 1.5mg/L,sodium deoxycholate 5g/L,and 5-bromo-4-chloro-3-indolyloctyl ester substrate concentration 0.2g/L.The growth rate of Salmonella on the test piece is higher than the quality control standard stipulated by the national standard.The specificity is good,and other non-target bacteria do not grow.The detection period is short,and the detection result can be obtained within 24 hours;The test piece has good stability and can be stored for 6 months at the temperature of 4℃;The test result of the test piece is accurate,and there is no significant difference between the growth rate of the test piece and the national standard culture medium plate(p>0.05).The detection results of actual samples are consistent with the national standard method.The coincidence rate of the detection results of food samples with the XLD plate of the national standard method is 98%,and that of BS plate is 96%.There is no significant difference in the detection results(p>0.05).The research results can speed up the detection of Salmonella and improve the detection efficiency,so it has high practical value.
