The special functional group to make special reaction which is formation of covalent chemical bonds,it is used to label and image of modified biomolecules in complex living system accurately or artificial regulation,meanwhile the two reactors don't reaction with the other's functional groups in organism,this kind of reaction is bioorthogonal reaction.The research of these kinds of methods are very important.So,these methods often are focused by Chemists and Biochemists.We develop a new type of [3+2] reaction which is strain-promoted and it is stimulated by UV.Particulartly,it is metalfree.How to find out the reactivity of the trans-Cycloheptenone and researching on this reaction.At the past we done research on total synthesis,we found out cycloheptenone which can do [4+2] reaction under UV.So we designed cycloheptenone to do [3+2] reaction under UV with azide compound,then we hope that it can work well with protein,this article introduce these next aspects:1,Making a presentation for history of development of bioorth-ogonal reaction and [3+2] reaction with azide compound.2,The mechanism of the reaction is cycloheptenone which become strained trans-cycloheptenone intermediate under UV,so it have a highly ring-strained,so it can do [3+2] reaction with azide compound.3,We have developed 7,7-two methyl cycloheptenone and a kind of azide compound which are most suitable.We also have synthesized thirty substrates of cycloheptenone and ten azide compounds.4,However cycloheptenone is a good acceptor of Michael reaction,so,we must develop a way to avoid the undesired reaction.We through literature review to find out a best way to deal with the problem.We find out that regulate pH can avoid the undesired reaction.So,we try to do this,finally we do successful.Then we have done the biorthogonal reaction.5,We also use BSA to be a model,which was used to do research on the biorthogonal reaction,meanwhile we are also success.So,we have realized remarking at biological macromolecule.This paper have developed a novel photo-controllable and metalfree biorthogonal reaction.The reaction is a new type of [3+2] reaction and cycloheptenone is cheap and easily got meanwhile it is stable in air.By the way,it is very useful.The paper have realized biorthogonal reaction of cycloheptenone and azide compound.
