Natural bioactive substances have the functions of promoting human health,preventing and treating diseases,but their bioavailability in human body is low.The delivery system of bioactive substances based on zein and polysaccharide has a good application prospect.In this study,zein was used as raw material and curcumin as model material.The interface between beet pectin(SBP)and cationic polysaccharide chitosan(CS)was prepared by using the method of anti-solvent.The protein polysaccharide interface composite particles and the stable Pickering emulsion delivery system were constructed to improve the loading capacity and biological activity of bioactive substances.Bioavailability of sexual substances.By measuring the particle size,potential,morphology and other basic properties of ZP / SBP cur in different proportion of SBP and zein,the loading ratio of composite colloidal particles to curcumin is relatively the highest when SBP: zein = 0.5:1 and 1:1,respectively,96.4% and 97.2%.At this time,the particle size of composite colloidal particles is 322.1nm and 322.7nm.Because SBP has adhesion to intestinal mucosa,simulated in vitro digestion shows that the system can prolong the residence time of curcumin in the digestive tract to 6h,provide a good time guarantee for the absorption of loaded nutrients in the intestinal tract,and improve the bioavailability.Curcumin content in the blood of Wister rats: Curcumin suspension and ZP cur composite particles reached the maximum concentration of curcumin in the blood at 20 minutes,which were 4.45ng/mL and 4.83ng/mL respectively,and then the concentration value decreased,consistent with the previous study.However,the ZP / SBP cur system prepared in this study still has a good protective effect on curcumin at 20 min.at this time,curcumin concentration in blood is 2.43 ng / mL,and 5.21 ng / mL curcumin is measured at 4h.At the same time,ZP / CS cur,which is a delivery system of curcumin loaded,was studied.The experimental results show that the loading capacity is less than ZP / SBP cur,and the suspension is easy to flocculate.A double loading curcumin Pickering emulsion with high loading capacity was prepared by using ZP/CS-cur as particle stabilizer and oil phase dissolving curcumin.It shows that the loading capacity of curcumin is super high,and the loading rate can reach 89.27% of the total amount of curcumin.The emulsion form is good,stable and durable.It can be put in the dark for 4 days or more for 60 days.The simulated digestion shows that cur is released completely in 360 min in the ordinary Pickering emulsion system,while in the Pickering emulsion system with double curcumin,the amount of zinc released at cur is 390 min.It has been reduced and released completely in 450 min,that is,the target emulsion system has good slow release characteristics,which plays a good protective role for curcumin,and provides more possibilities for improving the utilization rate of biological activity.
