Water-soluble conjugated polymer is a kind of conjugated system with large?-?*conjugated electron structure in main chain,and side chain modified hydrophilic functional group.Because of its remarkable fluorescence amplification effect,good water solubility and biocompatibility,it has attracted widespread attention.At present,water-soluble fluorescent polymers have been successfully applied to the detection of metal ions,biomolecules and cell imaging.This dissertation mainly focused on the application of water-soluble polythiophene derivative PT-PHO and poly?phenylene ethynylene?conjugated polymer PPE-O-PEG in metal ions detection and bioimaging.The specific research contents are as follows:1.Water-soluble polythiophene derivatives with phosphonic acids as the side chain groups were synthesized by FeCl3 oxidation polymerization.The organic compound of every step in the synthesis was characterized by 1H NMR and MS and the final conjugated polymer?PT-PHO?was also characterized by 1H NMR,FTIR and GPC.We find that PT-PHO has a better selectivity to Al3+in hexamethylenetetramine-HCl(0.1 mol L-1,pH=5)buffer solution.Under the optimal experimental conditions,PT-PHO has a wide linear range and a low detection limit for the detection of Al3+.Finally,PT-PHO was applied to the detection of Al3+content in traditional pasta and a satisfactory result was obtained in this part.2.We also find that PT-PHO can selectively detect Ca2+in NH3-NH4Cl(0.1 mol L-1,pH=9)buffer solution.Under the optimal experimental conditions,PT-PHO has a wide linear range and a low detection limit for the detection of Ca2+.In addition,PT-PHO was also used for the specifical lysosomes imaging in cells.The experimental results show that PT-PHO can detect the concentration of Ca2+in lysosomes in this part.3.Based on the interaction between PPE-O-PEG and Pt?IV?,the potential application of PPE-O-PEG as Pt?IV?carrier in tumor therapy is investigated in this part.In the experiment,PPE-O-PEG and chloroplatinic acid are used to obtain the coordination compound PPE-O-PEG-Pt,which is systematically characterized.Cell experiment results show that PPE-O-PEG has good biocompatibility and nuclear targeting,and can be used as a carrier of platinum ions to transport Pt?IV?to the nucleus of tumor cells.Cytotoxicity experiments show that obvious apoptosis could be observed after PPE-O-PEG-Pt was added for 24 h.These results suggest that PPE-O-PEG will have a promising application in tumor chemotherapy and imaging.
