Cyclodextrin?CD?and its derivatives could form a supramolecular system with drugs though non-covalent interaction,improving the water solubility and bioavailability of poorly soluble drug.Cyclodextrin-based stimuli-responsive assembly have became a research hotspot.The characteristics of stimulus responses make the assemblies performance easily controlled by factors such as pH,temperature,light,magnetic field strength,reducing compounds,enzymes,and other factors,achieving intelligent control of assembly structure,and achieving targeted release and timed release of drugs.In this work,two modified?-CDs were used to construct a pH-responsive supramolecular assembly,and the structure and properties of the supramolecular assembly were studied.A solid inclusion compound was prepared using 2,6-dimethyl-?-CD?M-?-CD?triptolide?TN?.The details are as follows:?1?pH-responsive supramolecular nanoparticles constructed by negatively charged sulfobutylether7-?-CD?SBE7-?-CD?and positively charged cationic surfactant benzyl dodecyl dimethyl ammonium bromide?BDBAB?.The assembly/disassembly behavior of the constructed nanoparticles was investigated by Ultraviolet-visible spectroscopy?UV-Vis?,zeta potential,dynamic light scattering?DLS?and transmission electron microscope?TEM?.The obtained supramolecular nanoparticles were subjected to model drug DOX loading.The pH-responsive release behavior of drug-loaded nanoparticles was studied by TEM and UV-Vis.?2?mono-?6-pentaethylenehexaamine?-?-CD was synthetized.using negatively charged anionic surfactant sodium dodecylbenzene sulfonate?SDBS?as a building block pH-responsive nanoparticle.The assembly/disassembly behavior of the constructed supramolecular nanoparticles were investigated by UV-Vis,zeta potential,dynamic light scattering?DLS?and transmission electron microscope?TEM?.The drug loading behavior of nanoparticles was studied using TEM and UV-Vis.?3?The anti-tumor activity of the above two drug-loaded supramolecular nanoparticles and their inhibitory activity on normal cells were studied by MTS experiments.?4?Prepared solid inclusion compound of TN and DM-?-CD by saturated aqueous solution method.the inclusion behavior of DM-?-CD to TN was investigated by UV-Vis,1H HMR,2D ROESY,FTIR,XRD and SEM.And the thermal stability and water solubility of the inclusion compound and in vitro release were further studied.
