Metal-organic frameworks(MOFs)are organic-inorganic hybrid materials with intra-molecular pores formed by self-assembly of organic ligands and metal ions or clusters through coordination bonds.Due to its porosity and large specific surface area,metal-organic frameworks have been widely used in the fields of gas adsorption and separation,catalysis,and controlled drug release.Because the pores of the metal-organic frame are open,this property is conducive to the loading of the drug,but it is also because of the open pores that it leads to the loss of the drug after loading.In order to further improve the application of metalorganic frameworks in the direction of drug carriers,some macromolecular compounds are usually selected to modify them to improve the rate of drug release.Coating the drug carrier with a polymer compound can not only reduce the loss of the drug,but also can feedback some changes in the physiological indicators of the body,thereby forming a targeted drug release system.Therefore,in this paper,N-isopropylacrylamide(NIPAAm),acrylic acid(AA),and sodium allyl sulfonate(ALS)are radically polymerized into nanogels.Novel polyacid-based metal organic framework synthesized by octamolybdate(TBA4MoO26),copper chloride and organic ligands(1,4-bis(1,2,4-triazol-1methyl)benzene,BBTZ)Coating was performed to prepare a composite responsive drug carrier.The specific research content is as follows:(1)Synthesis of a pH-responsive nanogel,using N-isopropylacryl(NIPAAm),acrylic acid(AA),and sodium allylsulfonate(ALS)through a free radical solution Polymerization to prepare nano-scale microspheres(nanogels).The test proves that the nanogel has excellent biocompatibility,and has good binding ability and coating effect with the polyacid-based metal-organic framework.It can form a double pH response drug release with the metal-organic framework to control drug release and the purpose of increasing the rate of drug release.(2)Synthesis of a polyacid-based metal-organic framework with pH response.Hydrothermal method was used for octamolybdate(TBA4MoO26),copper chloride and organic ligands(1,4-bis(1,2).,4-triazol-1methyl)benzene,BBTZ)Synthesis of a new type of porous polyacid-based metal organic framework.The test proved that the porous compound has a good loading rate for a series of anticancer drugs,and canquickly release the drug under acidic conditions,achieving the purpose of responsive release.In addition,the porous compound can generate ROS under laser irradiation,and has a certain photodynamic therapeutic effect.Chemo-photodynamic synergy was achieved in in vivo experiments.
