Preparation Of Cordyceps Militaris Polysaccharide Effervescent Tablets And Its Protective Effect On Acute Liver Injury Induced By CCl₄ In Mice

Cordyceps militaris?L.?Link has a variety of bioactive components.As one of its main bioactive components,Cordyceps militaris polysaccharide has pharmacological effects such as protecting liver and kidney,antioxidation,regulating immunity,etc.It can be widely used in the research and development of health food and tonic food,and has a broad development prospect.In this study,Cordyceps militaris polysaccharide was extracted from cultured Cordyceps militaris.After separation and purification by column chromatography,oral absorption and utilization of Cordyceps militaris polysaccharide were studied.Cordyceps militaris Polysaccharide was prepared into effervescent tablets,and the preparation prescription and technology of Cordyceps militaris polysaccharide effervescent tablets were studied,and the quality analysis and liver protection of the finished products were studied.Taking the extraction rate of crude polysaccharide of Cordyceps militaris as the index,the extraction method was selected,and the extraction temperature,extraction time and the ratio of material to liquid were optimized by orthogonal test on the basis of the previous single factor test.The optimal conditions of hot water extraction were obtained as follows:extraction temperature 90?,extraction time 1.5h,ratio of material to liquid 1:20?g/ml?.Cordyceps militaris crude polysaccharides were purified and separated by DEAE-52 and Sephadex G-100 column chromatography to obtain purified components?CMPn?with a polysaccharide content of 93.83%.After the polysaccharide of Cordyceps militaris was replaced by iodine,it was administrated to SD rats by gavage,and then the iodine content in blood and main organs was measured by ICP-MS to indirectly characterize the polysaccharide content of Cordyceps militaris.After oral administration of polysaccharide from Cordyceps militaris,the polysaccharide was quickly absorbed into the blood.After one hour,the absorption rate in the blood was the highest,reaching 5.71%.After oral administration for 3 hours,the distribution of Cordyceps militaris polysaccharide in different organs was different,and its content in liver was the highest.Taking the angle of repose,moisture absorption rate,disintegration time limit,appearance,etc.as the investigation indicators,the formulation of the preparation of Cordyceps militaris polysaccharide effervescent tablets was screened and the quality analysis of the finished product was conducted.It was determined that lactose was the filler and absolute ethanol was the binder.The preparation process is as follows:the acid-base mass ratio of the effervescent agent is 1.4:1?g/g?,the additive amount of the lubricant PEG 6000is 2%,and the addition amount of the sweetener stevioside is 3%.According to the best prescription,the appearance,weight difference,disintegration time limit,brittleness,hardness and other indexes of Cordyceps militaris polysaccharide effervescent tablets are all in accordance with the 2015 edition of the Chinese Pharmacopoeia.The average pH value of the solution is?4.71±0.05??RSD=1.04%?,and the content of polysaccharide is?9.52±0.09?%?RSD=0.95%?.The results of stability test showed that after 3,6,9 and 12 months,the appearance,disintegration time limit,pH and polysaccharide content of Cordyceps militaris polysaccharide effervescent tablets had no significant change,and the stability was good.Taking Kunming mice as experimental objects,the protective effect of Cordyceps militaris polysaccharide effervescent tablet on acute liver injury induced by CCl₄ in mice was explored.Compared with the model group,the activity of ALT and AST in the serum of mice in the Cordyceps militaris effervescent tablet group was significantly reduced?P<0.01?,the MDA content in liver homogenate was significantly reduced?P<0.05?,and the activity of SOD was significantly increased?P<0.05?.In the observation results of liver histopathology in mice,the pathological conditions such as liver cell necrosis and inflammatory cell infiltration in the effervescent tablet group were all reduced,which indicated that Cordyceps militaris polysaccharide effervescent tablets had a certain improvement effect on liver cell damage.
