In this paper,the interaction mechanism of Radix Polygoni Multiflori protein and curcumin was studied based on the materials of Radix Polygoni Multiflori protein,pectin and corn oil.Then,the function of the nanoparticles was evaluated,and the mechanism of the protein and pectin was studied.The factors affecting the interaction and the rheological properties under different factors were precisely controlled,so as to prepare high inward emulsion oil colloids.The system found a definite pH.as follows:(1)Protein delivery system: CAP,soybean protein as raw materials.The interaction between curcumin and scutellaria protein and soybean protein was studied.Steady state fluorescence analysis,Fourier transform infrared spectrum analysis,circular dichroism analysis,differential scanning calorimetry and thermal stability analysis of particle size and curcumin were used.The results showed that curcumin quenched the internal fluorescence of CAP and soybean protein in a static way,and the spontaneous reaction of Gibbs free energy reduction occurred in the hydrophobic interaction.Under the same conditions,the binding ability of curcumin to soybean protein was slightly higher than that of Baishouwu protein.Fourier transform infrared spectroscopy further confirmed the binding of curcumin and two proteins through hydrophobic interaction.Curcumin binding changed the secondary structure of the two proteins.The stability of curcumin during heat treatment can be effectively improved by the protein of CAP and soybean protein,but the protective effect of CAP is relatively weak.(2)Composite nano delivery system: with the CAP and pectin as materials,through heat treatment,the white Shouwu protein / pectin formed a soluble electrostatic complex at pH3.5to prepare nanoparticles.The results showed that compared with BSP / LPE nanoparticles,BSP/ HPE nanoparticles had smaller size,tighter structure,higher surface absolute potential and hydrophobicity.Curcumin is mainly embedded in the nano carrier through hydrogen bond and hydrophobic interaction.The nano composite can significantly enhance the optical stability of curcumin under UV irradiation.(3)Under the same pH and operating conditions,the particle size of the complexagglomerate reached the maximum value in the process of pH3 ~ 4.When the ratio of protein to pectin was 4:1,the complex agglomerate had better rheological property,and the high methoxylated pectin had more rheological property than the low methoxylated pectin Good rheological properties and better oil holding capacity lay the theoretical foundation for subsequent preparation of high internal emulsion.emulsion delivery system: high internal phase emulsion was prepared by high-pressure homogenization according to the complex coagulant,and the appearance was observed.The internal microscopic state was observed by optical microscopy and laser confocal microscopy,and the viscoelasticity of the high phase emulsion was further studied by rheology and thixotropic experiments.The structure shows that under suitable pH conditions,the emulsion has good stability and stable storage.After loading curcumin loaded with active substances,the physical properties of curcumin emulsion were studied.The experimental results showed that the rheological properties of curcumin loaded high internal emulsions were good,indicating that the gel network system was stable,curcumin resistance to irradiation in the emulsion increased the bioavailability of curcumin.
