Antibiotic resistance genes in the air invaded the body and caused potential harm to the organism.Atmospheric particulate matter is the carrier for antibiotic resistance genes in the air to enter the human body.Compared with PM2.5,PM1can enter the blood and cause more harm to human health.There are very few studies on atmospheric PM1carrying antibiotic resistance genes.In this study,the diversity,content and health risks of resistance genes in PM1in the air environment of Handan City were studied,and the microbial flora structure in PM1was analyzed.In this study,samples of PM1from Handan Environment Monitoring Center were selected and real-time fluorescent quantitative PCR was used to determine the contents of 16Sr RNA,int1,sul2,tet A,tet Q,erm C and aac C2 were measured by a real-time fluorescent quantitative PCR instrument.The concentrations were 2.21×105copies/m3,3.2×104copies/m3,2.82×102copies/m3,1.69×102copies/m3,5.51×102copies/m3,3.52×102copies/m3and 8.14×102copies/m3.The 16S r DNA amplicon sequencing technology was used to analyze the composition of microbiota in atmospheric PM1samples,including species annotation,?diversity,?diversity and function prediction.?1?The Venn diagram based on OTU analysis showed that the number of common OTUs in spring and autumn bacteria was the highest,indicating the highest similarity of OTU composition in spring and autumn;the relative abundance of species showed that at the genus level,the dominant bacterial genera in the four seasons were Stenotrophomonas;?2??diversity:Weighted Unifrac distance showed the greatest difference in species and abundance between summer and autumn bacteria;The Unweighted Unifrac distance showed the greatest difference in species composition between spring and autumn bacteria;The PCo A analysis based on the Weighted Unifrac distance showed the largest difference in species composition between spring and autumn;PCA analysis based on OTU level showed that the microbiota structure in spring and winter was the most similar;NMDS analysis based on OTU level showed that the spring and winter microbiota had the smallest differences;UPGMA clustering based on Weighted Unifrac and Unweighted Unifrac distances both showed that the difference in species diversity between autumn and winter microbiota is greatest;?3?The PICRUSt function prediction showed that the bacterial gene functions in the four seasons were similar,and there was no significant difference.This thesis clarified the pollution characteristics of antibiotic resistance genes in PM1in Handan City,revealed the types and contents of antibiotic resistance genes in PM1in the atmosphere,and conducted a health risk assessment of antibiotic resistance genes;explored the characteristics of the microbial communities in PM1,and the microbial community structure in the four seasons was compared and analyzed.Conducted a comparative analysis.The results of this study can provide a theoretical basis for the prevention and control of antibiotic resistance gene pollution in PM1in Handan City.
