The technology that separates circulating tumor cells from normal blood cells is called membrane filtration technology,which is based on the physical properties of circulating tumor cells and normal blood cells.The technique can be used to screen out tumor cells from a small amount of peripheral blood.It plays an important and positive role in early diagnosis,adjuvant diagnosis and treatment,diagnosis and prognosis,individualized treatment,rapid judgement of chemotherapy / radiation therapy,detection of individual drug resistance and monitoring of tumor recurrence.On the basis of membrane filtration and separation technology,the manufacturing process of microporous filter membrane is discussed first,and the specific cell enrichment test of microporous filter membrane is conducted.Secondly,according to the functional requirements of automatic membrane separation and circulating tumor cell device,we use related knowledge of electrical and electronics,mechanics,biology,and other disciplines,design and teste the separation,enrichment and dyeing system of circulating tumor cells based on negative pressure and multilayer composite membrane.A series of actions such as automatic sampling,separating,enriching and dyeing are completed by coordinating a series of motors and pumps and collecting multi-channel sensor signals.Besides,based on the idea of master-slave control,we use the STM32 chip and UCOS embedded operation system to carry out the circuit design of the lower computer system and the corresponding software development.For the host computer,based on the LCD screen and its control circuit board,built in embedded Linux operation system and QT4,the man-machine interaction interface is designed,and the program is programmed to complete the interaction function between the screen and the human.RS232 communication is used to achieve two-way communication with the lower computer.In the Fluent software,the separation process is simulated and analyzed based on the membrane separation process,and the stress situation of the microporous filter membrane is emphatically analyzed.In the liquid flow in the process of microporous membrane by blood components plug hole,leading to the porosity,causing the pressure into the microporous membrane under a reasonable interval,and even lead to the rupture of membrane blocked,a one-dimensional fuzzy control rules in this paper,through experimental verification,can make the pressure stable in a microporous membrane reasonable interval,the microporous membrane can completely blocked protective film is not broken down.Finally,the medical instrument Inspection Institute carried out full performance test,EMC/EMI test and passed according to the state regulations.The circulating tumor cell separation and dyeing equipment developed in this paper has high sampling accuracy and operation efficiency,and has a high degree of automation.It accords with the electrical requirements of the state for medical devices,and its performance is consistent with the original design requirements.
