Chicken-claw-shaped[Polyporus umbellatus(Pers.)Fries.]is the specific wild Polyporus species of Changbai mountains.It has been the medicine as a history of 2000 years in China.The life history was experiences basidiospore-mycelium-sclerotium-fruit body 4 times of the Chicken-claw-shaped.At present,the domestic main study of sclerotium pharmacological activities,etc,and fruiting body correlation studies are very few,this paper studies on the key problem of polyporus umbellatus fruitbody development.In the development process of chicken-claw-shaped polyporus fruiting body functional genes and metabolic regulation rules such as biological analysis theory basis.Experiments using chicken-claw-shaped polyporus fruiting body collected from Xie Fang Village,tian nan township,jiao he,jilin as test materials,and extracting total RNA through the Trizol improved method,building full length cDNA Library via SMARTTM cDNA Library Construction Kit.Building not amplified cDNA library by X TriplEx2 as a vector,and obtain the original library drops to 5.63 x 106,through restructuring at a rate of 98.73%,blue and white spot selection calculation purpose fragment mainly distributed in 500-2 000 bp.Drop degrees of the new library in 1.095 x 1010 pfu/mL.Database is about:2.19 x 1012 pfu,and selected 20 genes to sequence,will receive the cDNA fragments of pretreated with DNAstart software,length between 400-1 200 bp,are of high quality ests,ORF,5 'UTR region and 3' UTR region.Analysis of Blast in Nr/Nt database.The results show that successfully extracted total RNA from chicken-claw-shaped polyporus fruiting body,and builded the high quality full length cDNA library,for further research on growth mechanism of chicken-claw-shaped polyporus fruiting body providing the theoretical basis.
