The seeds of Bletilla Striata common in southern of Shaanxi were used as experiment material,the relationship between fruit form and seed setting rate,and the relationship between storage conditions and seed vitality were researched at first,addition compared the different effect of solid and liquid medium on seed germination,and further researched the effect of liquid medium components on seed development,addition researched the process of seed germination in liquid medium forming protocorms,and protocorms forming seedlings,preliminary established the technology system of the seed of B.Striata cultured in liquid medium quickly forming seedlings.The purpose of this study is to provide evidence and support for raising seedling technology of B.Striata seed.The results were as follows:1.The weight of fruit on the same infructescence showed most closely correlation with seeds setting rate,attaining intermediate degree.The correlation between diameter of fruit and seeds setting rate was higher than the length of fruit.And the weight of seeds in a fruit had a strong correlation with the weight of fruit(the correlation coefficient was 0.97).So the weight and diameter of fruit could serve as a standard to choose and evaluate the quality.2.Seeds treated with natural and silica gel drying were stored in different temperatures(5?,-18?,-80?).The vitality of seeds with natural drying stored in different duration(15,28,43,66,82 and 97 days)were evaluated,and it were found that the germination rate dropped to about 50% after 97 days.Meanwhile,the vitality of seeds with silica gel drying stored in different duration(15,43,and 97 days)was determined and the germination rate maintained highly,and showing that the lower the temperature,the higher germination rate was.So the silica gel drying method was better than the natural drying method to keep the seeds alive.3.The seeds of B.Striata were sowed in solid and liquid MS medium,and the diameter of protocorm and the length of leaf were measured during different times(14,28,35 and 42days).The results showed that the growth in solid medium was better than it in liquid medium,the length of leaf was 40% higher than that in liquid medium since 42 days,and the protocorm did not show obvious difference.So the germination condition of protocorm inliquid medium could achieve the desired effect.4.The seeds of B.Striata were cultured in vitro,using 10 different liquid media and deionized water,studying the effect of media composition on seed development.The results were: seeds germinated in all conditions;MS medium was the best medium for protocorm growth,followed by the 1/2 MS of inorganic,and it was the worst in 1/10 MS inorganic medium as well as in deionized water.The leaf began to grow significantly from 32 days with the radial growth of protocorm.5.The protocorms cultured in liquid medium for 45 days grow to seedling on the sterile soils,and the seedlings can be transplanted after 20 days and can grow normally.The result show that this set of cultivation methods established technical basis of B.Striata seeds cultured in liquid media germinating and grow to seedling.
