Nuclear factor Y(NF-Y)is a ubiquitous transcription factor involved in regulating important physiological and developmental processes.Previously,28 NF-Y genes have been identified in rice.5 OsNF-Y genes in our analysis were endosperm-specific gene,which belong to NF-YA,NF-YB and NF-YC,respectively.However,some of genes were not included in previous study.Therefore,this study intends to supplement and identify NF-Y family in rice genome.The analysis of expression data and co-expression gene of NF-Y family in rice will provide the basis for the study of OsNF-Y gene.Meanwhile,the function and regulation mechanism of endosperm-specific NF-Y gene was studied through the transgenic method,which has the application value to improve the yield and quality of rice.The main results are as follows:1.There were 34 OsNF-Y genes in rice;six of them were newly demonstrated members.All 34 OsNF-Y genes were dispersed on the 11 chromosomes(except chromosome 11),presenting an uneven distribution in all regions of the chromosomes.2.Expression profile analysis covering the whole life cycle of rice has revealed that the transcripts of OsNF-Y gene family differentially accumulated in a tissue-specific/preferential or constitutive manner.The OsNF-Y gene family contains seven pairs of segmental duplication gene and two pairs of tandem duplication gene.3.Nine OsNF-Y genes were differentially expressed after treatment with one or more of the abiotic stressors like drought,salt and cold in rice seedlings.4.Analysis of expression correlation and Gene Ontology annotation suggested that OsNF-Y genes were co-expressed with genes that participated in stress,seed storage reserves accumulation and plant development.Co-expression analysis also revealed that OsNF-Y genes might interact with each other,suggesting that the NF-Y subunit formed complexes that take part in transcriptional regulation.5.Real time PCR and detection of GFP fluorescence in transgenic plants revealed that OsNF-YC11 was endosperm-specific gene and has the highest expression value at 7 DAP.There was no obvious phenotype in the vegetative stage of OsNF-YC11 overexpression plants.The length of mature seeds was increased and the width of mature seeds was not significantly changed.6.The analysis of the content of starch and protein in the mature seeds revealed that the starch content of overexpression seeds was significantly decreased and the content of alkali soluble protein and alcohol soluble protein was increased.The expression of starch synthesis and storage protein related gene indicated that the expression of starch synthesis gene was down-regulated and the expression of storage protein related gene was up-regulated.These results suggested that OsNF-YCll could be involved in the accumulation of seed storage material.
