Rice has become an important model organism for the research of monocots because of its relatively small genome,efficient transformation techniques and extensive genetic resources.At present japonica rice have had a Nipponbare reference genome of high quality,however indica rice 93-11 genome sequence exists many problems.In this study,PacBio sequencing of ZS97 and MH63 genomes was implemented using clone-by-clone strategy,and an information platform was developed for storage,assembly and calibration of the sequencing results.The main contents are as follows:1)Preliminary constructed physical map was refined based on reference genome.The total number of clone contigs of ZS97 was reduced from 1736 to 539,the average coverage was 89.35%,and the total number of clone contigs of MH63 was reduced from 1083 to 401,the average coverage was 91.08%.For ZS97 and MH63,respectively,4,714 and 4,751 minimal tiling path BAC was selected from the physical map then sequenced,and 4,571 and 4,488 BAC sequences were obtained.2)The sequencing results of BAC sequences were assembled based on the physical map and reference genome.The size,number and N50 of genome contig sequence of ZS97 and MH63 is 347 Mbp,237,2,339,070 bp and 360 Mbp,181,3,097,358 bp,respectively.The function of genome assembling part of our web site was improved,including assigning contigs to chromosome,displaying basic information of contigs,marking filter sequence,generating and downloading AGP files and data verification.3)The completeness of the centromere sequences was analyzed.There were 2 and 4 complete centromere sequences detected in ZS97 and MH63.
