Plant secondary cell wall biosynthesis is regulated by various types of transcription factors.In Arabidopsis thaliana,a number of transcription factors have been identified to participate in secondary cell wall biosynthesis.However,very little is reported about the MYB transcription factors in rice.Hence,this study focused to characterize the OsMYB103L and related other MYB gene involved in the regulatory network of secondary cell wall biosynthesis in rice by using bioinformatics,genetics and biochemistry methods.The major results were described below:1.Total eight OsMYB genes were selected from OsMYB103L microarray data,and all genes exhibited high co-expression coefficients with the genes that are associated with cell wall biosynthesis under high expression levels at heading stages of rice;2.Overexpression and RNAi transgenic rice plants were generated using Agribacterium mediated transformation;3.RNAi plants of OsMYB86.1 exhibited rolled leaves phenotype towards abaxial side and reduced pollen vitality,and leaves of OsMYB61.2 RNAi plants also rolled towards abaxial side after heading stage;4.Polyclonal antibodies of OsMYB86.1 and OsMYB 103L were generated for further biochemical analysis,and both antibodies were examined with high specific recognition;5.ChIP-seq were performed with OsMYB 103L polyclonal antibody using the stem tissues of 1st,2nd and 3rd internodes,and the candidate genes that were directly regulated by OsMYB 103 L were identified.
