| In the current,cucumber fruit color was not only one of the most important commodity characters,but also the primary factor which had attracted more and more consumers.To study the inheritance of tender cucumber fruit peel color traits,this research used three inbred lines of cucumber which are distinct different in fruit color as the parents material,maked two hybrid combinations respectively and maked six genetic populations respectively.Through the screening SSR markers,we acquired the pleomorphic primers and constructed the cucumber genetic linkage map using the software QTL IciMapping.And we scanned the QTL associated with the cucumber fruit color.The main results were as follows.1.Maked six genetic populations and studied the genetic development about the tender fruit color in cucumber This research used three inbred lines of cucumber which are Q16(Dark green of fruit color)、Q8(Yellowish white of fruit color)and 24(Yellowish white of fruit color)as the parents material,maked six genetic populations respectively.The data of fruit colors were analyzed by joint analysis of multiple generations of the major gene plus poly-genes model of plant quantitative traits.the results showed the inheritance of fruit color in these two hybridized combinations(Q16xQ8 and Q16xQ24)were appropriate for the model of two major genes with additive-dominance-epitasis(model B-1).The heritability of major genes in two hybrid combinations was highest in F2 population which reached 87.4%and to 93.1%,and the environmental implication of major genes was relatively small.2.The screening of the pleomorphic primers in two hybrid combinations Through the screening of 995 pairs SSR markers,the cross of Q16 x Q8 produced 121 pairs polymorphism primers,the polymorphism rate was 12.2%.And the cross of Q16 x Q24 produced 126 pairs polymorphism primers,the polymorphism rate was 12.7%.We respectively analyzed 79 pairs and 80 pairs polymorphism primers in cross Q16 x Q8 and Q16xQ24.3.Constructed the genetic linkage map of two hybridized combinations We constructed the cucumber genetic linkage map of the cross Q16 x Q8 and Q16 x Q24.The genetic linkage map of the cross Q16 x Q8 contained 7 linkage groups and 74 SSR markers.The genetic linkage map of the cross Q16 x Q24 contained 7 linkage groups and 76 SSR markers.4.Preliminary QTL mapping of tender fruit color in cucumber We researched the QTL of tender fruit color in cucumber which based on the genetic linkage map.We found two QTL which located in LG1 and LG3 linkage group in cross Q16 x Q8.The locus on the LG1 was located between the primer SSR04079 and SSR03886,The site which on the LG3 linkage group was located between the primer SSR10370 and SSR06791.The phenotypic variation which can explain was 9.276%and 50.622%respectively.The cross Q16 x Q24 had one locus related to the cucumber fruit color on the LG3.This site was between SSR10370 and SSR05572,and the phenotypic variation which can explain was 68.976%. |
