Pine wood nematode(Bursaphelenchus xylophilus)is considered to be "pine cancer",is a very serious disease harm to pine.through parts of fujian were nematodes were nematodes and the hybrid,to explore the pathogenicity of crossbred,morphological changes,etc.,as a reference of pine wood nematode disease resistance breeding.This study proposed by fujian Yanping,Fuzhou forest park,and quanzhou anxi were nematodes of reciprocal cross,and Measured the hybrids and parents of the morphological data,detecting its pathogenesis and RAPD cluster analysis,and in the process of experiment validated by some of the test method,The results of the study summarized are as follows.1.Bursaphelenchus xylophilus with Bursaphelenchus mucronatus hybrids and their parents were pathogenic analysis,found that generally pathogenic can be divided into two categories,strong or weak,in this experiment were nematode pathogenic is relatively weak,were nematodes were nematodes and the hybrid offspring of pine trees are pathogenic,parents and some hybrids,such as C(nanping yanping district plans were nematodes ?x fuzhou forest park were nematodes came)pathogenic stronger case,were nematodes were nematodes and the hybrid,there will be a pathogenic stronger,should pay more attention to,what is the specific causes for the further study of the pathogenic stronger to be later.While other hybrids,although not in pathogenic stronger but basically has certain pathogenic effect.Level with the onset of value and the disease index of pine wood nematode were reported in two kinds of indexes to describe the degree,and the test results are close to two indicators,the difference is't large.2.nematodes were nematodes and the hybrids and their parents to cluster analysis,the form can be divided into three classes of 10 kinds of nematodes.After hybridization in the part of the nematodes needle mouth,esophagus,genital malformation etc.3.Selected 14 primers on hybrids and their parents of RAPD cluster effect is obvious,as a whole the average 70%higher polymorphic primers,amplify the stripes are clear and complete can also use this article 14 primers used to identify were nematodes were nematodes and the two kinds.Hybrids RAPD cluster can be divided into four categories:(1)the yeonpyeong area plans were nematodes separate category;(2)B(nanping yeonpyeong district plans were nematodes ?x minqing were nematodes came),C(nanping yeonpyeong district plans were nematodes ?x fuzhou forest park were nematodes came),D(nanping yeonpyeong district plans were nematodes came by fuzhou forest park were nematode ?),E(nanping yeonpyeong district plans were nematodes ?xquanzhou anxi were nematodes came)4 samples to;(3)were nematodes parent,nanping jianou,forest park,fuzhou,quanzhou anxi three sample for a category;(4)F(nanping yeonpyeong district plans were nematodes came by quanzhou anxi were nematodes ?)to separate the last category.Combined with the conclusion above,It is concluded that the pathogenicity of pine wood nematode and form relationship more closely,by RAPD clustering can also get the classification of similar results.
