To investigate the mechanism of fat deposition in Huai pig,we choose leptin(LEP)and leptin receptor(LEPR)genes,which are related to the performance of fat deposition.Using paraffin-section method to observe the morphological characteristics of adipose tissue,and to analyze the morphological indexes of fat bubbles.Using real-time quantitative PCR method to detect the gene expression difference of LEP and LEPR between breeds,different breeds,tissues and periods,and we took study the expression law of fat deposition,analysis of correlation between the performance of the pig fat deposition.1 Morphological analysis of adipose tissues between Huai pig and Duroc.This study shows that the shape of fat bubble of Huai pig was irregular oval,and the differences between short diameters of fat bubble of Duroc were not significant.The diameter,area,density,average diameter of fat bubble of Huai pig were significantly difference with Duroc(P<0.05).Compared to Duroc,the fat bubble of Huai pig is more hypertrophic and development earlier.The morphological differences between different adipose tissues become smaller with body maturation.In the same pig breed,the fat tissue of back,abdominal fat and suet,coronary artery had significant differences(P<0.05),and there was more fat deposition in subcutaneous than visceral.2 Expression of LEP and LEPR genes between Huai pig and Duroc.This study shows that expression of LEP and LEPR genes of adipose tissues of Huai and Duroc have differences,which regulate fat metabolism and maintain energy balance,but their expression were reverse regulation.In five adipose tissues,LEP gene expression highest in backfat,followed is the up backfat,lower abdominal fat and suet,in the coronary fat was the lowest.LEP gene expression of subcutaneous adipose was significantly higher than visceral fat(P<0.05).LEP gene expression of Huai pig was significantly higher than Duroc(P<0.05).The mRNA expression of LEP gene of adipose tissues in 7-month-old Huai pig had significant difference from 12-month-old Huai pig and 7-month-old Duroc.The majority of the gene expression of LEP and LEPR showed positive correlation.However,7-month-old Duroc and Huai pig was not significant in coronary(P>0.05).Compared with the 12-month-old Huai pig,only the mRNA expression difference of LEPR gene of arterial fat in 7-month-old Huai pig was significant(P<0.05).The expression of LEP and LEPR genes were higher in tissues,which contain more adipose.
