Taxus chinensis var.mairei(Lemee et Levl)Cheng et L.K.Fu is a variation of Taxaceae Taxus in china,and it contains Taxol,which is a diterpenoid of taxane with excellent activity against cancers,in its branch,bark,needle,root and pulp.However,Taxus plants grow slowly,and yield relatively low amounts of paclitaxel,all of this cause supply crisis of paclitaxel.Cell cultures and fungal fermentation of Taxus spp.,have rich supply of paclitaxel and related taxanes,this technology is viewed as a potential alternative to producing paclitaxel.Systematically summarizes about the research of new studies at home and abroad in several aspects,such as the bionomics,pharmacognosy research,chemical composition,pharmacologic action,reproductive technology,genetic engineering,cell culture,fungus fermentation.Further--more,discusses the problems of the biotechnology research.Macroscopic and microscopic observation were used to authenticate this crude drug,and the identification characteristics were studied.To establish methods for identification of the shoots,leaves and roots of Taxus chinesis var.mairei.The leaf of Taxus chinensis var·Mairei,Stomatal bands,tracheid and andcrystal fibre can be regarded as identification characters.The 10-DAB provides a basis for TLC identification.Optimize the extraction process of Taxus chinensis mairei.In this article,an orthogonal test wasf conducted,with 5 major terpenoids.The five terpenoids were compared by RP-HPLC method.The results showed that the optimal extraction process was adding 6-times ethanol of 85%concentration,and extracting for twice,2 hours each time.The factors which influence the Taxus chinensis var.mairei callus induction and growth rate were studied by a single factor analysis and the orthogonal experiment design.The five taxane diterpenoids including paclitaxel in callus from different sources were determined by the HPLC analysis.The weighted score for each the above factors mentioned would be taken account to optimise the tissue culture conditions.The most optimum culture medium of the callus induction was B5+2,4-D 3.0 mg·L-1+6-BA 1.0 mg·L-1+NAA 1.0 mg·L-1+KT 0.5 mg·L-1.To investigate effects of the extracts from the branches and leaves of Taxus chinensis var.mairei and callus on alpha-glycosidase activity,was determined by Bemfeld,PNPG and GOD.The results showed that extracts from the branches and leaves of Taxus chinensis var.mairei and callus all has activity against alpha-glycosidase,and callus is better.Preliminary selection of five medicinal fungi were selected,which were based on previous research experience,and combined with the characteristics of strain growth itself.four kinds of medicinal fungi were selected for the fermentation of the leaves and roots of Taxus chinesis var.mairei,such as Ganoderma lucidum.The results showed that four medicinal fungi selected could grow better in the leaves and roots matrix.Besides,there were effects of different degree on the contents between different types of taxane diterpenoids.The fermenting condition of culture optimizing study for Ganoderma lucidum-Taxus chinesis var.mairei using of RSM.In this article,an orthogonal test wasf conducted,with 5 major terpenoids.The most optimum fermenting condition was adding 6-times water,PH,culture temperature,sample particle size.At the different time as research object,investigated through analyzing amylase,glucoamylase,cellulase lipase and protease enzyme activity,wtih effects oftaxane diterpenoids terminal point of fermentation was determined on the 34-41th day.Analysis of Inositol about six medicinal fungal fermentation and tissue culture of Taxus chinesis var.Mairei by GC-MS.The results showed that the Inositol in tender branches and leaves high than olders,VA content higher than other fermentation group,seed samples 4 higher than other callus.
