Rice grassy stunt virus(RGSV),negative sense ssRNA virus,is transmitted by the brown planthopper(BPH)in a persistent-propagative manner.RGSV has caused serious damage to rice plants in South,South-east,and East Asia.RGSV infecting causes leaf yellowing,stunting,excess tillering and brown or rust-colored sports on rice leaves.Among which,excess tillering is the typical symptom of RGSV infecting.Experiment shows that,compared with WT,BPH is more likely to eat RGSV infecting rice.RGSV P3 protein is disease-related protein and relates with OsJM1.In this study,we got OsJMT1 transgenic plants through agrobacterium-mediated method.Then,we detect the eating behaviors of BPH between WT and transgenic plants.Y2H and BIFC are also used to detect the interaction between P3 and OsJMT1.We are aimed to find out how P3 protein affects BPH eating behavior and the function of OsJMTl in this process.The results we got are as below.1.We found that BPH is more likely to eat in P3 transgenic rice compared with WT.But the eating behavior of BPH doesn't have much difference between OsJMT1 transgenic rice and WT.2.We know that RGSV P3 interact with OsJMT1.So we detected the residue of P3 which play an important role in this interaction.The experiment was revealed that residue between 124 and 195 played an essential role in this interaction.The second structure of this domain encoded protein is a Beta strand.3.The cellular localization of P3 and OsJMT1 encoded proteins is in the plasma membrane and the nucleus.Those results have laid the foundation for the understanding of the functions of RGSV P3 protein and the differences between BPH eating behaviors.
