The flowering time of bamboo plants is uncertainty,and most of the bamboo will into death after flowering.Phyllostachys violascens is one of the main shoot bamboo used as vegetable in northwest zhejiang,bamboo shoot yield decreased and some of them died after flowering,could cause economic losses.Flower is the sign of higher plants into reproductive growth,are essential for plants to complete the life cycle.Flowering of plants regulating by a variety of internal factors and external environmental factors,that regulation by the complex and strict genetic network.Discovery of Key flowering gene and network research in Arabidops is,provides an excellent reference to study of bamboo flowering induction mechanism.In this studty,we isolated PvJ3 from Phyllostachys violascens,and studied the function of it,the research results are as following :1 、 Using homology cloning method,a J3 homolog named PvJ3 was isolated from Phyllostachys violascens,which is explore the functions of J3 gene in bamboo plants.The full-length ORF of PvJ3 is 1260 bp,which encodes 419 amino acids.Structure analysis of PvJ3 protein shows that the protein is a typical DnaJ domain,so it’s one of the J family.The PvJ3 showed highly homology with other species through sequencing alignment and phylogenetic analys is.Through the prediction physicochemical property and hydrophobic/hydrophilic of PvJ3 protein,the results show that the protein iis strong solubility and poor stability.2、Real-time quantitative PCR results showed that the PvJ3 was expressed in all the organs both in flowering and non-flowering bamboo plants,especially the highest expression in stem and the lowest expression in the leaves of flowering bamboo plants;In the non-flowering bamboo,also the highest expression in the stem,low expression in young and mature leaves.3、Protein subcellular localization showed that the PvJ3 located in cytoplasm and cell nucleus.4、The phenotype of overexpression of PvJ3 in Arabidopsis showed that predate flowering and more main stems.5、Bimolecular Fluorescence Complementation results showed that PvJ3 protein can interact with PvSVP and PvFT1,and localization in the nucleus.6、Yeast two hybrid results showed that PvJ3 protein can interact with PvSVP and PvFT1,which consistent with BiFC.And prove the C end of PvJ3 is functional domains.7、Yeast two hybrid results showed that PvJ3 protein can interact with PvFT1/PvFT2/ PvFT4 and PvFT5,and not interact with PvFT3.Suggest that PvJ3 protein selective interact with PvFT1-PvFT5.
