Cyclophilin A(CypA)is one of the members of Cyclophilin protein family with cis-trans isomerases Peptidyl-prolyl activity,and plays an important role in protein folding,apoptosis,virous infection and inflammation.Cyclophilin A,which is widely distributed in eukaryotes and prokaryotes.However,the exact role of CypA in insect innate immunity remains undetermined.Here,we will study whether CypA participates in inhibiting host cell immune in Spodoptera Litura which was dominated by MbB V.Here,we found that host hemacyte apoptosis was increased after parasitization by microplitis bicoloratus.Meanwhile,the expression of CypA have a significant increase.So we hypothesized that CypA may be involved in the hemacyte apoptosis.CypA is highly expressed in Hi5 cells infected by MbBV,Which resulted in cell apoptosis.When CypA was silenced by SiRNA-CypA in Hi5 cells,cell apoptosis was markedly reduced.Furthermore,we found that CypA and AIF were co located in nucleaus from cytoplasm in infected cells,which caused cell DNA ladder.While CypA was suppressed by CsA,nuclear distribution of AIF was blocked.In conclusion,we found that MbBV may be upregulated CypA in immunosuppression of Spodoptera litura,which assisted with AIF nuclear distribution from cytoplasm,then resulted in genome DNA fragmaents.So,hemacytes happened to apoptosis and cell immune of Spodoptera litura was inhibited by MbBV in parasitization.
