[Objective]:Fluorosis is widespread and its metabolites are mainly accumulated in the kidneys.This thesis studies the effects of exogenous calcium on fluorosis in rats exposed to fluoride after renal injury and its mechanism,lay a theoretical foundation for the fluorosis relief mechanism to further elucidate the mechanism of fluoride on kidney injury and calcium,and provide an important theoretical basis for the implementation of scientific diagnosis and intervention for fluoride poisoning.[Methods]:160 SD rats of 4 weeks old size were randomly divided into 5 groups:control group,fluoride group,fluoride deficiency group,calcium fluoride group and calcium supplement group.The control group of rats(C)fed with normal diet and drinking distilled water.Fluoride group rats(F)fed with normal diet and drinking water fluoride(150mg/L)fluoride supplementation group,low calcium fluoride,calcium fluoride group fill fill high calcium sodium fluoride in drinking water(150mg/L)fed with feed containing respectively.0.5%CaCO3,1%CaCO3,2%CaC03.The feeding,drinking and body weight of rats was observed and recorded during the feeding period.Experiments 30d,60d,90d,120d.At the end of the experiment,the rats were enucleation and the neck broken.Renal tissue was collected,endoparasitic reticulum stress related genes and proteins were measured,morphological structure was observed,Real-time,PCR and Western-blot techniques were used to detect the expression levels of related genes mRNA and protein[Results]:1 HE staining showed that the kidney of rats exposed to fluoride was injured,and the kidneys were slightly damaged after calcium addition.2 Real-time PCR study found that fluoride can make the net stress pathway of rat ER related gene expression is abnormal,and lead to Askl,Traf2,Irel,Mkk7,Caspase-3,Caspase-9 genes;Caspase-12,Jnk gene expression was up-regulated,indicating differences in expression of adding calcium can greatly the degree of balance caused by fluorine.The results of the 3 Western-blots experiment showed that the intake of fluoride increased in expression levels of caspase-12 and Jnk proteins,while the addition of calcium alleviated the extent of the expression level to some extent.[Conclusion]:cause kidney damage in rats with excess fluoride poisoning,injury was significantly improved after the addition of calcium,fluoride poisoning can cause Ask 1,Traf2,Ire 1,Mkk7,Caspase-3,Caspase-9 genes;Caspase-12,Jnk gene expression was significantly up-regulated,adding calcium can antagonize the toxicity of fluoride on kidney injury.
