Tibetan cashmere goat,a domestic goat breed in the northwestern area of Tibet,have a great economic value.At present,the production of cashmere,mutton and milk can' t meet the needs of people in Tibetan cashmere goat because of their low fecundity.Therefore,the most efficient way to improve the fecundity of Tibetan cashmere goat is that how to choose the candidate genes of high fecundity.Previous studies showed that BMP15,GDF9 and BMPR1B were strongly involved in early follicular growth and development.But little is known about the association between these three genes and fecundity in Tibetan cashmere goat.213 female Tibetan cashmere goats were chosen and polymorphisms of BMP15,GDF9 and BMPR1B were analysed with PCR-RFLP and sequencing.The main results obtained were as follows:1.Coding sequences length of BMP 15 and GDF9 were 1185 bp and 1362 bp,respectively.BMP 15 and GDF9 were highly similar(99%)with Jining Grey Goat and Tibetan cashmere goat shared homologous with Capra Hircus and Ovis Aries.2.Two new SNP loci were detected in BMP15(G732A and C805G).G-A mutation at G732A didn't change amino acid while the C805G changed amino acid(Q-E).Genotypes GG,GA,AA were detected at G732A and allele frequencies were 0.695,0.282 and 0.023 respectively.Genotypes CC,CG,GG at C805G were detected and allele frequencies were 0.620,0.320 and 0.060 respectively.3.Two published SNP loci(C719T and G1189A)of GDF9 were detected in Tibetan cashmere goat and both of them were led to the amino acid change(A-V at C719T,V-I at G1189A).Genotypes CC,CT(TT wasn't detected)were detected at C719T locus and allele frequencies were 0.994 and 0.056,respectively.Genotypes GG,GA and AA were detected at G1189A locus and allele frequencies were 0.579,0.305 and 0.056,respectively.Two published SNP loci(G3 and G4)of GDF9 were homozygous mutation in Tibetan cashmere goat.4.Mutation loci of G1,B2,B3,B4,FecXH,FecX1,FecXL,G2,G5,G6,G7,G8,FecGE,FecTT and FecB didn't exist in Tibetan cashmere goat.Results indicated that Tibetan cashmere goat,other goat and sheep breeds shared high similarity and they were homologous.BMP15,GDF9 and BMPR1B showed genetic conservation in Tibetan cashmere goat.Polymorphism distribution of BMP15,GDF9 and BMPR1B were similar among Tibetan cashmere goat,Yimeng black goat and other goat breeds.This study could add basic data to the selection of high fecundity breed in Tibetan cashmere goat.
